摘　　要：采用酵母表达载体进行重组人α８型干扰素（ＨｕＩＦＮα８）的表达，该表达菌株可将重组α８型干扰素分泌进入培养基中，回收培养基进行纯化，然后利用超滤浓缩，先过Ｓ．ＳｅｐｈａｒｏｓｅＦＦ柱，再进行ＤＥＡＥＳｅｐｈａｒｏｓｅＦＦ层析和Ｇ２５脱盐，最后用高效液相ＨＰＬＣ，ＳＤＳＰＡＧＥ丙烯酰胺凝胶电泳及肽图等方法对纯化产品进行鉴定。采用以上两步纯化方法纯化酵母分泌表达的重组人α８型干扰素纯度可达９５％以上，其比活性为３０×１０８ｕ／ｍｇ。The recombinant human interferon α 8(IFN α 8) is expressed by yeast expression system.The expression strain can secreted recombinant interferon α 8 into culture.The culture containing interferon α 8 is ultrafiltrated,concentrated and chromatographed with S Sepphrose FF,DEAE Sepharose FF.The treated sample is desalted by G 25 .Then the pure sample is identified by HPLC,SDS PAGE electrophoresis and peptide map.Two steps purification can obtain high pure interforon α 8 .The pure is higher than 95%.The specific antiviral activity of interforon α 8 is 3.1×10 8 IU/mg protein.
