网织红细胞计数质控物的研制

摘　　要：本室采取给兔注射苯肼,人为使兔贫血,获得富含网织红细胞(reticulocyte,Ret)的兔血。经7mmol/L中性红活体染色后,用0.5％戊二醛固定细胞,试制Ret计数质控物。质控物制片后,用14mmol/L中性红复染,常规Ret计数质控。本文质控物标定值为:RBC(x±2S)=(2.65±0.15)×10^(12)/L,CV=2.8％;Ret(x±2S)=(169.80±42.40×10~9/L,CV=12.5％。经6个月稳定性观察结果显示,此质控物4℃保存半年,稳定性好,适用于常规计数Ret的室内室间质量评价。Blood specimens with abundant reticulocytes obtained from anemic rabbits induced by injection of phengylhydrazin were stained with 7 mmol/L neutral red and fixed successively by 0. 5% glutaraldehyde. Then the smears were re-stained by 14 mmol/L neutral red and used as the quality-control material ofr reticulocyte count. The parameters of this material were as follows: RBC= (2. 56± 0. 15)×1010/L, CV=2. 8%; Ret=(169. 80+42. 40)×1010/L, CV = 12. 5%.Experiments on this material showed that this material still maintained high stability at 4℃ 6 mouths later, which suggests that this material is highly stable and can be used in the internal and external quality assessment in reticulocyte count.
