人胎盘催乳素的纯化与鉴定

摘　　要：以正常产妇的新鲜胎盘为原料，经过盐析、等电点沉淀、离心等粗提，再经ＤＥＡＥ─纤维素阴离子交换层析，Ｓｅｐｈａｄｅｘ－Ｇ－１００凝胶过滤，并首次利用圆盘制备电泳纯化。最后获得人胎盘催乳素纯品，其活性采用１２５Ｉ－ｈＰＬ放免检测药盒测定。Human placental Lactogen (HPL) was isolated from fresh normal human placenta by salting ,precipitating at isoelectric point and ultracentrifugating. The purification was performed by three steps. They are anionexchange chromatography of DEAE-cellulose , sephadex -G - 100 gel filtration and preparative disc electrophoresis. Each step was tested for its immunoactivity by RIA of radiolabelled 125I-HPL.
