摘 要:^(60)Coγ线6.5Gy照射小鼠经每天2次、连续4天皮下注射500μg/kg的重组人白细胞介素6(rhIL-6) 时,10天后照射小鼠脾结节数(CFU-S)、脾重和造血祖细胞数(CFC-Mix、CFC-GM和CFC-MK)明显增高,但红系造血祖细胞数(CFC-E)轻度降低。另外在照后14-22天期间,外周血白细胞、红细胞及血小板数明显增高。进一步研究还发现,rhIL-6明显提高照射小鼠13天的脾结节(CFU-S)、骨髓CFU-Mix的自杀率,因而提示rhIL-6能刺激照射小鼠造血干细胞的增殖作用。结果提示,当在照射小鼠体内应用rhIL-6时,rhIL-6能刺激多能造血干细胞向粒系、红系和巨核系祖细胞分化,因而是一种有效的多向性造血生长因子且可望用于临床。Recombinant human interleukin-6 (rhIL-6) was administered s.c. twice a day at a daily dose of 500μg/ kg body weight to 6.5 Gy 60Co γ rays irradiated mice for four days. After 10 days the number of spleen colony forming unit (CFU-S), spleen weight and number of progenitor celis (CFU-Mix, CFU-GM, and CFU-MK )were significantly increased but the number of CFC-E was slightly decreased in rhIL-6-treated mice. The number of cells in peripheral blood, including white blood celi, red blood celi and platelet were increased significantly between days 14 to 22 after irradiation. In further studies, suicide rates of CFU-S and CFU-Mix were all much higher in rhIL-6-treated mice than those in saline-treated control mice, indicating that rhIL-6 could stimulate the proliferation of hematopoietic stem celis in bone marrow of irradiated mice.These results suggested that rhIL-6 is an effective stimulator of multipotent hematopoietic stem celis to differentiate into myeloid, erythroid, and thrombocytic lineages when administered in vivo to irradiated mice and indicated a possible therapeutic potential of rhIL-6 in clinical situations.
