摘 要:应用干细胞培养及集落形成法观察到白细胞介素11(IL-11)虽不能单独使小鼠骨髓细胞形成集落,但和白细胞介素3(IL-3)合用时,对正常及5-FU处理后小鼠骨髓细胞的集落形成有明显的促进作用,其中含巨核细胞的集落(CFI-GMM+CFU-M)数较IL-3单独组分别增加约2.5和7倍。巨核细胞的体积测量法显示IL-11可增大成熟巨核细胞的体积。复种实验表明巨核细胞的早期集落形成受IL-3的刺激,而进一步分化和增殖需要IL-11的参与。在干细胞因子(SCF)的协同下,IL-11可使5-FU处理后小鼠骨髓细胞形成多量的未分化集落(CFU-Blast),提示IL-11在造血干细胞早期的自我更新中可能起着重要的作用。Interleukin-11 (IL-11) is a recently cloned lymphohemopoietic cytokine. It was identified in medium conditioned by a cell line derived from primate bone marrow stromal cells (PU-34). IL-11 mainly regulates the function of T lymphocytes. On hematopoiesis, IL-11 supports IL-3-dependent proliferation of primitive progenitors and shortens the G0period of stem cells. In order to detect its effect on megakaryocytopoiesis, we used normal and 5-FU treated mouse bone marrow cells as target cells. IL-11 and IL-6, as two synergistic factors, were combined with IL-3 in multiple combinations. Serum-free culture was applied in all tests except for the concentration-response. We also tested the synergism of stem cell factor (SCF), a newly cloned hematopoietic factor, and 1L-11 on supporting the growth of early stem cells derived from 5-FU treated mouse bone marrow. The results show that when with IL-3, IL-11 could support more megakaryocyte colonies (CFU-GMM+CFU-M) than IL-3 alone did. The increases were 3.5 and 7-fold on normal and 5-FU treated cells respectively (P< 0.001). This effect was similar to that of IL-6. IL-11 also stimulated CFU-GM growth on IL-3-dependent progenitors in serum-free culture. Using megakaryocyte progenitors supported by IL-3 and IL-11, we found that IL-11 could enhance the cell size of megakaryocyte. In replanting test, IL-11 did not promote any colony growth in serum-free culture, while with IL-3, it stimulated more megakaryocyte colonies than IL-3 alone, although the total colony numbers were similar (P<0.01). On 5-FU treated cells IL-11 synergized with SCF and promoted the colony growth of considerable CPU-Blast. These results reveal that IL-11, like IL-6, is a potent megakaryocytopoietic factor and an important early stem cell growth factor as well.
