川芎注射液抗血小板作用机理的研究

摘　　要：应用体外实验方法观察主要含挥发油成分的川芎注射液对不同激活剂诱导的兔血小板聚集、血栓素 A_2(TXA_2)生成、Ca^(2+)内流及静止血小板环磷酸腺苷(cAMP)水平的影响。结果,川芎注射液对二磷酸腺苷(ADP)、花生四烯酸(AA)、血小板活化因子(PAF)、A_(23187)诱导的 TXA_2生成均有明显抑制作用,对 AA、ADD 诱导的血小板聚集有较强的抑制作用,对 PAF 诱导的血小板聚集无显著影响;提示川芎注射液主要通过抑制 TXA_2生成抑制血小板活化。川芎注射液能显著抑制 A_(23187)诱导的血小板聚集。提示其对血小板活化过程中胞浆游离钙迅速升高后环节有一定影响。川芎注射液对血小板外 Ca^(2+)内流无直接影响。川芎注射液能显著提高静止血小板 cAMP 水平。认为川芎注射液以抑制 TXA_2生成为主在多个环节上抑制血小板活化。The effects of Ligusticum wallichii injection(LWI)on rabbits' platelet aggregation,thromboxane A_2 (TXA_2)formation and Ca^(2+) influx induced by different stimuli and cAMP levels in non-activated platelets were observed in vitro.The results were as follows:(1)LWI significantly inhibited TXA_2 formation induced by adenosine diphosphate(ADP),arachidonic acid(AA),platelet activating factor(PAF)and A23187.It also sig- nificantly inhibited platelet aggregation induced by ADP and AA,which was dependent on AA metabolism:but it did not inhibit platelet aggregation induced by PAF,which was not dependent on AA metabolism.These re- sults suggested that LWI inhibited platelet activation through suppressing AA metabolism.(2)LWI significantly inhibited platelet aggregation induced by calcium ionophore A23187.This result indicated that LWI might have some effects on the phases after Ca^(2+) was raised in the course of platelet activation.(3)LWI had no significant effect to the platelet Ca^(2+) influx induced by PAF,but significantly inhibited that induced by AA.It suggested that LWI had no direct effect to the facilitated Ca^(2+) influx on platelet membrane.(4)LWI raised the cAMP lev- els in non-activated platelet.In conclusion.the LWI inhibited platelet activation in some different ways.but mainly through suppressing the AA metabolism.
