摘 要:用脑组织匀浆及尼龙网逐级过滤技术分离鼠脑微血管,再用胶原酶及胰酶处理使血管细胞分散后进行细胞培养,脑微血管内皮细胞Ⅷ因子相关抗原,γ-谷氨酰转肽酶及碱性磷酸酶染色均为阳性,光镜与电镜检查均显示为微血管内皮细胞结构。研究了7种生物肽对培养的内皮细胞的影响:它们都未能使细胞γ-谷氨酰转肽酶活力发生变化,对细胞的增殖效应也无统计学意义。但血管活性肠肽、神经降压素及强啡肽使细胞内cAMP含量提高(P<0.05),提示这3种生物肽在调节脑微血管内皮细胞正常生理功能上可能起一定作用。Microvessels were obtained by successive pasages of mouse brain homogenates through graded nylon meshes. Endothelial cells were then isolated for culture by treatmet with collagenase and trypsin.Endothelian cells were identified by positive tests with Factor Ⅷ-related antigen,γ-glutamyl transpeptidase and alkaline phosphatase, and also by light and electron microscopy.The effects of men-enkephalin,β-endorphin, dynomhin, neurotensin (NT), vasopressin, somatostatin and vasoactive intestinal peptide(VIP)on endothelial cells in culture were studied using cAMP, cGMP γ-GTP and acid phosphotase as indices.It was found that VIP, NT and dynorphin induced elevation of cAMP levels (P<0. 05).Suggesting these biopeptides may play an important role in the modulation of cerebro microvascular endothelial cell function.
