摘 要:应用反转录-聚合酶链式反应(reversetranscription-polymerasechainreaction,RT-PCR)方法,测定出一氧化氮合成酶(NitricOxideSynthase,NOS)mRNA不仅在心血管系统细胞中广泛存在,并且发现内皮素可以促进血管平滑肌细胞中NOSmRNA的表达(+34.8%),增高血管平滑肌细胞中NOS酶活性(+74%)。结果提示,由内皮素引起的血管平滑肌细胞中NOS基因表达和NOS活性的增加,可能是高血压中的一个重要的负反馈因素。ith reverse transcription polymerase chain reaction(RT-PCR),it had been found that NOS mRNA is widely distributed in cells of cardiovascular system such as cardiocvtes,endothelial cells,VSMCs, When ET(10- mol/L) was added to cultured VSMC,it significantly increased the NOS mRNA level in VSMC( +34.8%) .By measuring the enzyrme activity of NOS in cultured VSMC,it is foumd that when VSMC was treated with ET(2xl0-7 mol/L,NOS activation is significantly increased(79%)compared to the control.These da-ta showed that the increased expression of NOS gene in VSMC induced by ET may be an important regulatory factor in hypertension.
