无刺番麻组织培养快速繁殖种苗的研究

摘　　要：实现无刺番麻快速繁殖有两条途径：其一是芽长芽途径，即以吸芽的切块为起始材料，用ＭＳ＋ＢＡ３—５ｍｇ／Ｌ培养基，使腋芽和顶芽萌动生长，再用ＭＳ＋ＢＡ７ｍｇ／Ｌ＋蔗糖７％培养基继代培养，使芽不断增殖。培养３５天，繁殖系数达４．８。其二是胚胎发生途径，即以胚苗的叶片为起始材料，用ＭＳ＋２，４－Ｄ６ｍｇ／Ｌ＋ＮＡＡｌｍｇ／Ｌ＋ＢＡ４ｍｇ／Ｌ＋蔗搪７％培养基诱导愈伤组织，然后在ＭＳ＋ＮＡＡ０．１－０．２ｍｇ／Ｌ＋ＢＡｌ－２ｍｇ／Ｌ＋活性炭０．１％＋蔗糖５％的分化培养基上诱导胚状体的大量产生和萌发成苗。用组织培养方法繁殖无刺番麻，具有快速、高效、实用特点。Two procedures for rapid propagation of the thornless blue aloe havebeen developed,The explants were excised from suckers,sterilized andcultivated on Murashige and skoog(MS)basal medium containing3-5mg/LBA. The established axillary buds and terminal buds were subculturedon MS basal medium containing 7mg/L BA and 7% sucrose for budproliferation,The propagation index was 4.8 after 35 days culture.Another way of propagation is based on embryogeny,Aseptic leaves wereexcised and cultured on the Ms basal medium involving 6mg/L2,4 D,lmg/L NAA,4mg/L BA and 7% sucrose for caelus induction, and thentransferred onto differentiation cuIture medium of MS basal compositionembracing 0.1-0.2mg/L NAA,1-2mg/L BA,0.1% activated cqarbon and 5% surcose to induce extensive multiple embryoids to form plantlets.The plantlets were transferred into a sand bed mixed with coir dust(1:1,v/v)for acclimition,the survival rate being95% or more.
