氯硝柳胺控释剂药物动力学的研究

摘　　要：通过对氯硝柳胺控释剂这一双层(速释层和缓释层)包衣丸剂药物动力学的研究,观察到其降解过程是按一级反应进行,并得出有关药物动力学参数,该控释剂半减期长,具有缓慢释药的长效作用。现场施药及室内流水冲洗1年、1.5年后,该制剂尚剩余药量7.62％和19.5％,其浸液仍有灭螺效果;远比常规氯硝柳胺糊剂喷洒法的理论持效时间长(9.7d),理论用药量少。不仅避免药物浪费,而且减轻环境污染和对鱼类的毒害等。结果提示:氯硝柳胺控释剂是一个研制成功的新型长效杀螺剂。Pharmacokinetics of double layer coated pill (quick-release layer and slow-release one) of controlled-release formulation of niclosamide was examined. The parameters indicated that the decomposition of the formulation belonged to first class reaction with long half life and could release drug sustainedly with effective concentration for a long period. Field and laboratory trials confirmed that after the formulation washed by running water for 1 or 1. 5 years. it still retained 7. 62% and 19. 5% of the drug respectively and its extract maintained the effect of killing snail for much longer than 9. 7 days, the theoretical effective period of the paste form of niclosamide lasted, it was applied in spraying process. The new formulation not only can greatly save the original drug-base but also reduce the pollution to environment and the poison to fishes etc. The results demonstrated that the controlled-release formulation of niclosamide developed in our laboratory will be a promising molluscicide in schistosomiasis control program especially in complicated environments.
