用CFU-GM Do值推断人体辐射敏感性

摘  要:本文研究了动物与人体CFU-GM辐射敏感性与整体辐射敏感性的关系。结果表明,给小鼠全身1次照射不同剂量后取骨髓培养CFU-GM,与体外直接照射骨髓细胞的结果相比较,它们的D_O值分别为1.33Gy和1.23Gy,二者十分相近。通过对小鼠与人体各个个体CFU-GM辐射敏感性的测定,它们的D_O值差异很大,最大值与最低值相差2倍多。在慢性粒细胞白血病病人经全身^(60)Coγ—射线照射治疗的经过中,以及在骨髓移植的病例中,都观察到CFU-GM D_0值的大小能反映出全身照射后的辐射效应。It is known that radiation can produce radiation damage and sickness. The degree of this damage depends not only on radiation dose mainly, but also on the radiosensitivity of whole body. The different radiosensitivity exerts obvious influence on the effectiveness of cancer radiotherapy. The preconditioning radiation dose is very important to the success of bone marrow transplantation. Therefore, it is desirable to know individual radiosensitivity exactly before transplantation. This paper describes CFU-GM radiosensitivity of mouse, dog, monkey and human bone marrow cells in vitro and the method of predicting the radiosensitivity of whole body. CFU-GM radiosensitivity was expressed as CFU-GM D0 determined from the exponential part of survival curve. It was shown that the D0 value of bone marrow cells from whole body irradiated mice was 1.34 Gy, and 1.23 Gy when the cells were irradiated in vitro. The difference between these two D0 was not significant.This shows that the radiosensitivity in vitro may be substituted for the radiosensitivity to whole body irradiation. The radiosensitivity of CFU-GM from different individuals was various; for example, the D0 value was 0.84 ±0.045 Gy (0.53-1.27 Gy) for normal human CFU-GM and 0.59-1.78Gy for the CFU-GM from patients with leukemia and lymphoma. Different mouse individuals showed various D0 values of 0.65-2.01 Gy. The average values of mouse, dog, monkey and human were 1.23,0.57,0.63, and 0.79 Gy, respectively. These D0 were related to their LD50 for whole body irradiation. The radiosensitivity of whole body could thus be predicted from the CFU-GM D0 value determined in vitro. Our results show that D0 value of CFU-GM in vitro bears relation to whole body radiosensitivity.
