川芎丹参煎剂灌胃大鼠后体内川芎嗪的药物动力学研究

摘　　要：本文用反相高效液相色谱法,测定川芎丹参(每毫升含川芎生药3g、丹参生药1g)汤剂中川芎嗪含量平均为463.02μg/g·ml。川芎丹参煎剂一次灌胃(40g/kg)大鼠后体内川芎嗪的药物动力学参数如下:α2.3283±0.7192(h^(h^-1)),β0.4789±0.2893(h^(-1)),K_a 11.5080±2.8210(h^(-1)),K_(10)0.7380±0.2546(h^(-1)),K_(12)0.2054±0.0917(h^(-1)),K_(21)1.5109±0.4709(h^(-1)),t_(1/2)α 0.2977±0.2231(h),t1/2 β1.4473±0.9014(h),AUC 0.8357±0.1675(μg·h·ml^(-1)),V_(C/F7.3903±3.9889(L/kg)。Serum levels of Tetramethylpyrazine(TMP) were determined in six healthy rats after administering a single dose(40 g/kg) of Ligusticum Chuanxiong Hort and Radix Salviae Miltiorrhizae Decoction (LCH&RSMD) by HPLC method and pharmacokinetics of TMP was studied. The contents of TMP in LCH&RSMD is 463.02 μg/g·ml. Serum TMP was qualitatively analysed by HPLC and GC-MS. Pharmacokinetic parameters is as fallows: α 2.3283±0.7192(h-1), β 0.4789±0.2893(h-1), Ka 11.5080±2.82100(h-1), t1/2 α 0.2977±0.2231(h), t1/2 β 1.4473±0.9014(h), AUC 0.8357±0.1675(μg. h. ml-1), V. C/F 7.3903±3.9889(L/kg).
