摘 要:本文应用肾小球细胞体外培养方法和双同位素标记技术,观察了阿霉素肾病大鼠肾小球系膜细胞(GMC)对白细胞介素6的反应、肾病大鼠GMC条件培养液(GMC-CM)对正常大鼠GEC掺人^(35)S和~3H-亮氨酸的影响以及GMC-CM对小鼠骨髓瘤杂交瘤细胞增殖的作用。结果显示:①肾病大鼠GMC对不同浓度IL-6的反应曲线与正常GMC明显不同;②肾病极期GMC-CM明显促进正常大鼠GEC合成含硫化合物;③肾病大鼠GMC-CM中含有骨髓瘤细胞的杀伤物质。以上结果提示,阿霉素肾病大鼠GMC的部分生物活性发生了改变。Adrimycin (ADR)-treated rats have been used commonly as an animal model for the study of minimal change nephrotic syndrome. To identify whether or what changes of mesangial cells occurred in this ADR rats, methods of glomerular cell culture and dual-isotope labelling were used in this study. We investigated: ① the responsiveness of glomerular mesangial cell (GMC) in ADR rat; ② the effect of conditional medium of ADR rat GMC (OMC-CM) on S, H-Leucin uptake by nomal rat glomerular epithelial cell (GEC) and on the proliferation of mice myelohybrama MH60. The result showed that; ①the responsiveness of ADR rat GMC to IL-6 was different from the normal rats;② GMC-CM from ADR nephrotic rat increased significantly the synthesis of sulfated compounds by normal rat GEC;③there was some cytotoxic factor to MH60 in nephrotic rat GMC-CM. The results indicated that part biological function of GMC in ADR nephrotic rat had been changed.
