摘 要:描述了单子山楂的离体繁殖方法.结果表明,BGS 培养基有利于扦插枝条侧芽的萌发.MS 基本培养基加入0.1 mg·L^(-1)的 IBA 和1 mg·L^(-1)的 BAP 就能有效地诱导芽丛分化,再提高 BAP 的浓度或加入 GA_3都没有增加分化芽效.在诱导单子山楂枝条在培养基中生根时,NAA 的效果显著优于 IBA,其中尤以 NAA 0.5 mg·L^(-1)的用量最佳.两种基本培养基(1/2MS 和 MS)的生根效果差异不显著.连续黑暗处理一周不仅未增加枝条的生根率,反而使生根率显著降低.叶片愈伤组织诱导的初步研究表明,当加入0.1 mg·L^(-1)IBA 时,1~2 mg·L^(-1)的 BAP 能够有效地诱导愈伤组织产生.In vitro propagation of Crataegus monogyna has been carried out.The results showed that BGS medium was beneficial to germination of the lateral buds on stem segments from outdoor trees.Shoot proliferation was effectively induced on MS medium containing 0.1 mg·L^(-1) IBA and 1.0 mg·L^(-1) BAP. Further increase of BAP concentration or the addition of GA_s did not increase the number of proliferating shoots.The effect of NAA on the induction of root from in vitro shoots was significantly better than that of IBA,the best concentration of NAA was 0.5 mg.L^(-1).The effect of MS medium on root induction from in vitro shoots was not significantly different from 1/2 MS.Continuous darkness decreased the rooting percentage of in vitro shoots.The preliminary results also showed that MS medium,containing 0.1 mg· L'IBA and 1~2 mg·L^(-1) BAP,could effectively induced the production of calli from leaves of C. monogyna .
