A STUDY ON MICROPROPAGATION TECHNIQUE IN TISSUE CULTURE OF LARIX HYBRIDS

摘  要:利用13年生落叶松杂种休眠顶芽作外植体,采用腋芽增殖途径,分化率最高可达80.6%,由一个外植体进行一次分化培养最多可形成30个腋芽,经继代培养后多数能伸长形成嫩枝,少部分试管苗生根形成了完整的植株。较好的激素组合为BA3+NAA0.2;BA1+NAA0.01;BA2+NAA0.2(单位:mg/L)。细胞分裂素以BA分化效果最好,KT次之。同一杂种不同单株再生个体的能力有很大差别,较好的单株由一个外植体平构形成11.9个腋芽。Dormant buds of Larix hybrids of 13 year old were cultured in vitro. Auxiliary buds were formed on the explants and the largest differentiaton rate was 80.6%. The maximum number of auxiliary bud was 30 in one induction explant and most of them elongated into shoots. A few roots were formed on the shoots. The desirable hormone compositions for the auxiliary induction were BA3+NAA0.2; BA1+NAA0.01; BA2+NAA0.2(mg/l) and BA was more efficient than KT. There were significant differences in respect of the bud-forming capacity among individual plant of the same hybrid. On the average, 11.9 auxiliary buds were formed from the best plant.
