摘 要:以籼、粳稻及其杂种的成熟种胚愈伤组织胚性细胞系为材料,用NaCl作为选择剂,采取正选择法进行筛选。比较了不同基因型及不同筛选方法的选择效果,明确了耐盐细胞系的有效选择周期和其再生植株分化的主要影响因子,获得了耐盐的突变体。这些变异体脱离选择剂6个月后,耐性稳定,具有对盐逆境的系列适应性状,其再生植株后代及其幼穗培养也表现出耐盐性。Our research interest is being focused on the relationship between plant NaCl-resistance and the conditions inducing the variances of NaCl- resistance. This experiment was conducted to isolate and select the NaCl-resistant variants from the callus embryonic cell lines, derived from the mature seed embryos of ten cultivars or hybrids in Oryza sativa Var. hsien, O. sativa Var keng and their hybrids (see table 1), using positive selection, and to study their phys iological and biochemical charactreistics of the expression of resistance to NaCl and the resistant response to NaCl on the levels of both callus cultures and regenerated plants. In past four years, variants tolerant to 0. 5%-1. 0% NaCl solution were selected, which maintained the stable tolerance to NaCl solution after 6 months free from selection pressure, and showed a series of NaCl-tolerant adaptative characters. Furthermore, the progenies from the tolerant variants were also tolerant to NaCl.
