摘　　要：目的建立反相高效液相色谱法检测SD大鼠血清中坎地沙坦的含量。方法以萘普生为内标,按内标法定量。血清样品经盐酸酸化后,加入乙醚,离心分离,取上清液置于离心管中,水浴蒸干后用流动相溶解进样。色谱柱:依利特Hypersil ODS2 C18柱(4.6 mm×250 mm,5μm),流速:1 ml/min;柱温:25℃;流动相:甲醇-水-磷酸(77∶23∶0.1),紫外检测器的波长为252 nm。结果坎地沙坦和萘普生的保留时间分别为4.2和5.1 min,大鼠血清中坎地沙坦浓度在2.0～200.0μg/L范围内呈良好线性关系,R2=0.998 9。日内、日间精密度RSD均小于7%。低、中、高3个浓度样品的方法回收率和提取回收率分别为96.7%、98.0%、99.5%和71.0%、69.8%、70.4%,且样品稳定性良好。结论本法操作简便,灵敏度高,重复性好,可用于血清中坎地沙坦的含量测定。Objective To establish a method for determination of candesartan in SD rats blood serum by RP-HPLC. Methods Naprosyn was the internal standard material, according internal standard method to quantify the samples. After the samples were acidized by hydrochloric acid, these were extracted from serum with diethyl ether, then centrifugalized the samples. Liquid superuatant were assumed into the centrifuge tubes, and the centrifuge tubes were put into water bath, the liquid supernatant were evaporated to dryness, then the remains were dissolved with mobile phase and detected by RP-HPLC. Chromatography was performed on an Hypersil ODS2 Cl8 column （4.6 mm ×250 mm, 5 μm） with a mobile phase consisting of methanol -water -phosphoric acid （77 ： 23 ： 0.1 ） at a flow rate of 1.0 mL/min. The temperature of column was 25 ℃. Quantification was performed by UV detection at 252 nm. Results The peak time by HPLC of candesatan, naprosyn were 4.2, 5.1 min, respectively. The standard curve was linear with a concentration range of 2.0 - 200.0 μg/L with R2 = 0. 9989. The relative standard deviations （RSD） of intra-day and inter-day were 〈 7%. The method recovery and extraction recovery at low, medium, high concentration were 96.7% ,98.0% ,99.5% and 71.0% ,69.8% ,70.4%, respectively. And the stability of samples were very stable. Conclusion The method was found to be simple, sensitive, accurate and reproducible for determination of candesartan in rats blood serum.
