摘  要:目的建立止咳水中盐酸氯胺酮、氢溴酸右美沙芬及盐酸曲马多的HPLC-MS/MS筛查方法。方法采用Eclipse XDB C18色谱柱(4.6 mm×50 mm,5μm);流动相:乙腈-0.4%乙酸铵溶液(内含0.2%三乙胺,用冰醋酸调pH值至5.5)(35∶65);流速:0.8 ml/min(分流比1∶4);柱温:35℃;串联四极杆质谱,ESI(+)电离,采用MRM监测模式进行测试。结果实现了3个物质的同时测定,分离度好,峰形佳;检测限(DL):盐酸氯胺酮1.0 ng,氢溴酸右美沙芬0.5 ng,盐酸曲马多0.5 ng。结论本方法灵敏、简便、可靠,可应用于止咳水中盐酸曲马多及盐酸氯胺酮的非法添加筛查,或者应用于鉴别止咳水中的主要成分氢溴酸右美沙芬。To establish a HPLC-MS/MS method for detecting of ketamine hydrochloride,dextromethorphan hydrobromide and tramadol hydrochloride in cough mixture. Methods Eclipse XDB C18 column (4.6 mm ×50 mm, 5 μm) was applied, the mobile phase was a mixture of acetonitrile -0.4% ammonium acetate solution ( containing 0.2% triethylamine, acetic adjusted to pH value 5.5 ) ( 35:65 ), the flow rate was 0.8 ml/min ( split ratio 1 : 4 ), and the temperature was set at 35 ℃ ; triple quadrupole mass spectrometry with ESI ( + ) and MRM mode. Results Enables up to 3 simultaneous determination of a substance, separation, and LOD: ketamine hydrochloride was 1.0 ng and dextromethorphan hydrobromide was 0.5 ng, tramadol hydrochloride was 0.5 ng. Conclusion This sensitive method is simple and reliable. It can be applied to detecting illegally added ketamine hydrochloride and tramadol hydrochloride, or identifving dextromethorohan hvdrobromide in cough mixture.
