摘 要:[目的]检测CD4、CD25、CD127在佐剂关节炎(AA)大鼠胸腺、脾脏中的表达情况及新风胶囊(XFC)对其的影响,探讨AA大鼠免疫功能改变机制。[方法]将60只Wistar雄性大鼠随机分为正常对照组、模型对照组和新风胶囊、甲氨喋呤、雷公藤多苷片3个治疗组,每组12只,用弗氏完全佐剂诱发大鼠产生关节炎(正常对照组除外),测量大鼠体重、足趾肿胀度,计算关节炎指数、胸腺指数、脾脏指数;采用免疫组化SP法,检测各组大鼠胸腺、脾脏中CD4、CD25、CD127的表达情况。[结果]与正常对照组相比,模型对照组大鼠胸腺指数、脾脏指数差异显著,CD4、CD25及CD127阳性表达率均有所下降;与模型对照组相比,治疗组均能改善大鼠胸腺、脾脏指数,升高CD4、CD25及CD127阳性表达率;各治疗组相比,在升高CD4、CD25及CD127阳性表达率方面,XFC组优于其余治疗组,在改善AA大鼠胸腺、脾脏指数方面,各组相比无显著差异(P>0.05)。[结论]AA大鼠的关节病变与胸腺、脾脏指数升高,胸腺及脾脏中CD4、CD25及CD127表达减少,调节性T细胞分泌不足导致免疫复合物沉积增多有关;XFC可以降低胸腺及脾脏指数,升高胸腺及脾脏中CD4、CD25及CD127的表达水平,减少免疫复合物的沉积,从而改善AA大鼠的关节病变,此为XFC改善AA大鼠关节病变的作用机制之一。[Objective] To dectect the expression of CD4, CD25, CD127 in rats thymus, spleen with adjuvant arthritis(adjuvant arthritis, AA), analyze the influence of XFC on this alteration and explore the mechanisms of the variation of immune function in rats with AA. [Methods] Induced the arthritis in rats with Freund' s complete adjuvant(CFA), then measured the body weight and swollen toes of rats with AA,calculated the arthritis index,the index of thymus and spleen;Immunohistochemical SP methods was adopted to detect the expression of CD4,CD25 and CD127 in thymus, spleen of normal group(12cases)and AA rats(48cases);observed the morphology of thymus, spleen with electron microscopy. [Results] Compared with normal group, there were significant differences in thymus index, spleen index of AA rats;the positive rate of CD4,CD25 and CD127 in the thymus, spleen had declined; the ultrastructure of thymus and spleen of model group was damaged. [Conclusion] The degree of joint disease of AA rats was in relation to the increasing thymus and spleen index, declined CD4, CD25 and CD127 expression, and the decreasing Treg. XFC could lower the thymus and spleen index, rise the CD4,CD25 and CD127 expression, reduce the immune complex to improve the joint disease,it may be one of the mechanisms of XFC to improve tha joint disease of AA rats.
