二甲双胍联合吡格列酮对肥胖糖尿病大鼠胰岛β细胞的保护作用

摘  要:目的 观察二甲双胍联合吡格列酮对肥胖糖尿病(DM)大鼠胰岛β细胞的保护.方法 腹腔注射链脲佐菌素30mg/kg,制成糖尿病动物模型.50只健康Wistar雄性大鼠随机分为5组:正常对照组和糖尿病组给予等量蒸馏水,二甲双胍治疗组给予300mg·kg-1·d-1溶于蒸馏水中灌胃,吡格列酮治疗组给予10mg·kg-1·d-1溶于蒸馏水中灌胃,二甲双胍+吡格列酮治疗组给予二甲双胍300mg·kg-1·d-1和吡格列酮10mg·kg-1·d-1溶于蒸馏水中灌胃,干预12周.观察治疗后5组DM大鼠空腹血糖、糖化血红蛋白(HbA1c)、血脂、空腹胰岛素水平并进行比较.结果 与正常对照组比较,DM对照组大鼠FPG,FINS,TC,TG,HbA1C及Homa-IR均明显升高(P<0.01),Homa-β明显下降(P<0.01);二甲双胍组与DM对照组比较,大鼠FPG,FINS,TC,HbA1C及Homa-IR均明显下降(P<0.01或P<0.01),Homa-β明显升高(P<0.01),而TG无明显降低(P>0.05);吡格列酮与DM对照组比较,大鼠FPG,FINS,TG,HbA1C及Homa-IR均明显下降(P<0.01),Homa-β明显升高(P<0.01),TC无明显降低(P>0.05);二甲双胍组和吡格列酮组FPG,HbA1C均较DM对照组明显降低,但两组间比较差异无显著性(P>0.05);吡格列酮组和二甲双胍组比较FINS,Homa-IR明显减低,Homa-β明显升高(P<0.01);二甲双胍联合吡格列酮组与吡格列酮组比较,大鼠FPG,FINS,TC,HbA1C及Homa-IR均明显下降(P<0.05),Homa-β明显升高(P<0.01),差异有统计学意义.结论 盐酸吡格列酮联合二甲双胍对肥胖糖尿病大鼠胰岛细胞的保护作用优于单用上述两种药物.Objective To exlore the protection of pioglitazone combined with metformin for islet β cell of rat with diabetes mellitus(DM). Methods Intraperitoneal injection of streptnzotocin(30mg/kg) was used to establish diabetic animal models. Fifty healthy male Wistar rats were randomly divided into 5 groups,in which,the rats in the control group and diahetes group were given distilled water and metformin(300mg·kg-1·d-1 ,orally) dissolved in it,the rats in the pioglitazone treatment group received solution garage( lOmg·kg-1·d-1 in distilled water, and the rats in metformin + pioglitazone treatment group received metformin ( 300mg·kg-1·d-1 ) and pioglitazone ( lOmg ·kg-1·d-1) dissolved in distilled water for 12 weeks. After the treatment, fasting plasma glucose( FPG), glycosylated hemoglobin ( HbAI c) ,blnod lipids and fasting insulin levels of diabetic rats in all groups were detected and compared. Results Compared with the control group,FPG, FINS,TC,TG, HbAI C and Homa-IR of diabetic control rats were significantly inereased( P 〈 0.01) ,while the Homa-β was decreased significantly( P 〈 O. Ol ) ; in mefformin group and dial)etic control group, rats FPG, FINS, TC, HbAI C and Homa-IR were significantly decreased ( P 〈 O. O1 ) , Hnma-β significantly increased ( P 〈 O. 01 ) , while no significant decrease in TG ( P 〉 O. 05 ) ; cmnpared with diabetic control group, the level of FPG, FINS, TG, HbAI C and Homa-IR in pinglitazone group were significantly decreased ( P 〈 O. O1 ) , while Homa-β significantly increased( P 〈 O. 01) ,TC had no significant decrease(P 〉 0.05) ;in metformin group and pioglitaznne group, the level of FPG and HbAI C was significantly decreased than those with diabetes,but the difference was not significant( P 〉0.05) ;compared with the metformin group,the level of FINS and Homa-IR was significantly reduced,and Homa-β significantly increased in pioglitazone group( P 〈 O. 01 ) ; compared with pioglitazon
