摘 要:目的探讨用硅胶环预置法建立新型小鼠全层皮肤切除创面模型作为研究创面愈合机制、评价药物疗效的工具。方法在小鼠背部皮下左侧预置硅胶环,1周后固定组硅胶环再次以间断缝合固定于皮肤,非固定组硅胶环与皮肤不予固定。于植入硅胶环中心部位及右侧对应位置建立全层皮肤缺损后,通过大体观察及组织学检测分析比较各组创面的愈合。结果硅胶环固定组与非固定组、对照组比较,收缩性愈合比重减低(P〈0.05),上皮化、增生性愈合修复比例增高(P〈0.05),非固定组与对照组间差异无统计学意义(P〉0.05)。结论硅胶环预置法较旧的小鼠创面愈合模型可更好模拟人类创面修复模式。Objective Provide a novel, reproducible, reliable murine excisonal wound model by implanting a silicone ring subcutaneously in advance of dorsal wound creation. Methods A silicone ring was implanted subcutaneously in one slide of the mouse dorsum one week before further operation. In sutured group (n = 12), silicone rings were sutured with surrounding normal skin, whereas rings remained untouched in unsntured group (n = 12). 8 mm full-thickness excisional wounds were created both in the center of silicon ring and on the other side of midline (control group, n = 12). Wounds were observed and evaluated, epidermal thickness, collagen architecture and deposition, cell proliferation and angiogenesis were assessed. Results Compared to control group or unsutured group, wound healing in sutured group was significantly prolonged ( P 〈 0. 05 ). Wound re-epithelialization and collagen deposition were found im- proved in fixed group( P 〈 0.05 ). No significant difference was found between control group and unfixed group( P 〉 0. 05 ). Conclusion The excisional wound mode/with advanced silicone ring implantation can mimic and reflect the procedure of human wound healing better when compared with existing models. It can be considered as an ideal model for further study of wound healing.
