摘  要:目的观察一氧化氮(NO)供体L-精氨酸(L-Arg)对豚鼠耳蜗外毛细胞L型钙通道电流(ICa-L)的影响。方法以40只杂色或白色豚鼠为研究对象,采用急性酶分离方法分离豚鼠耳蜗外毛细胞,根据溶液中加入的L-Arg浓度分为0.5×10-6 mol/L组(A组)、1.0×10-6 mol/L组(B组)、1.5×10-6 mol/L组(C组),采用全细胞膜片钳技术分别记录不同浓度L-Arg组外毛细胞L-型钙通道电流。结果低浓度的NO供体L-Arg可以抑制ICa-L,此作用具有电压依赖性,对反转电位无明显影响。在指令电压0mV时,峰值电流密度增幅最大值A组为-2.12%,用药前后差异无统计学意义(P>0.05);B组增幅为-30.69%,用药前后差异有统计学意义(P<0.01);C组增幅为-65.08%,用药前后差异有统计学意义(P<0.01)。结论低浓度的NO可以抑制ICa-L。Objective To evaluate the impact of nitric oxide(NO) donor L-arginine(L-Arg) on the L-type calcium channel current(ICa-L) of cochlear outer hair cells in guinea pig.Methods Forty mottled or white guinea pigs were used for the study.Acute enzymatic separation method was used to isolate outer hair cells of guinea pig.Based on the L-Arg concentration,0.5 × 10-6mol / L group(A group),1.0 × 10-6mol / L group(B group) and 1.5 × 10-6mol / L group(C group) were separated.Whole cell patch clamp technique was used to record the effects of L-Arg on L-type calcium channel current of outer hair cell with different concentrations.Results Low concentrations of L-Arg inhibited the ICa-L,which was a voltage-dependent and had no effect on the reversal potential.When the command voltage of 0mV,the maximum increase in peak current density was-2.12% in group A with no significant difference between before and after treatment(P 0.05);B group increased by-30.69% with statistical significance(P0.01) between before and after treatment;C group increased by-65.08%,with significantly difference(P0.01) between before and after treatment.Conclusion Low concentrations of NO can inhibit the ICa-L.
