摘  要:目的 探讨IL-1β对RAW264.7细胞表达IL-4,GATA-3mRNA的影响.方法 常规培养RAW264.7细胞后进行干预,A组:IL-1β(0mg/L,1mg/L,10mg/L,20mg/L)刺激细胞1h,B组:IL-1β(10mg/L)刺激0h,30min,1h,3h,6h,C组:IL-1β(10mg/L)刺激0h,30min,1h,1.5h,2h,采用反转录聚合酶链反应(RT-PCR)分别检测IL-4,GATA-3mRNA的表达.结果 IL-Iβ刺激RAW264.7细胞不同浓度、不同时间与空白对照组比较IL-4,GATA-3mRNA的表达增高(P<0.05);IL-1β(10mg/L)刺激RAW264.7细胞1h时,IL-4mRNA的表达量最高,1.5h时GATA-3mRNA的表达量最高.结论 IL-1β在RAW264.7细胞中促进IL-4,GATA-3mRNA的表达呈浓度、时间依赖性,提示IL-1β通过促进单核巨噬细胞分泌IL-4,GATA-3,诱导Th2型细胞分化,促进和加重哮喘的慢性炎症.Objective To observe the effect of IL-1β on expressions of IL-4,GATA-3mRNA of RAW264.7 cells. Methods The cultured cells of the fourth to fifth passage were used for experiments. 2 ×10^5/ml cells were seeded in 24-well plates,when cells reached 80% filsion,the following treatment would he clone, group A : cells were treated with IL-1β ( 0mg/L, 1 mg/L, 10mg/L, 20mg/L) for 1 h, group B : cells were treated with IL-1β(10mg/L) for 0h,30min,lh,3h,6h,group C:cells were treated with IL-IiS(10rag/L) for 0h,30min,1h,1.5h, 2h. Expressiun of IL-4 and GATA-3mRNA in group A,expression of IL-4mRNA in group B and expression of GATA-3mRNA in group C were detected by reverse transcription polymerase chain reaction( RT-PCR). Results Increase of IL-4 and GATA-3mRNA expression was found when RAW264.7 cells were stimulated with different concentrations and different time compared with the control group ( P 〈 0.05 ). When RAW264.7 cells were treated with IL-1β(10mg/L) for 1 h, expression of IL-4mRNA reached the highest level;When treated for 1.5h,expression level of GATA-3mRNA was the highest. Conclusion IL-113 induces IL-4 and GATA-3mRNA expression iu a time-dependent and concentration-dependent manner in RAW264.7 cells,which suggests that IL-1β induces the monocyte macrophages production of IL-4 and GA- TA-3,induces differentiation of Th2 type cells,and plays an important role in promoting and aggravating chronic inflammation of asthma.
