摘  要:经过NaCl溶液匀浆抽提、硫酸铵分级分离、SP-SepharoseFastFlow层析、SephacrylS-100层析、Macro-Cap-SP层析,从苦瓜籽中同时分离纯化出大量的α-苦瓜素和苦瓜抗HIV蛋白30。经SDS-PAGE鉴定,二者均为单一条带,表观分子量为28kDa。用两种蛋白分别免疫Balb/c小鼠,双向免疫扩散表明,两种苦瓜籽蛋白无交叉反应。本实验为进一步探索PEG修饰MAP30免疫原性变化打下了基础。Large amounts of α-momorcharin(α-MMC) and MAP30 were quickly and easily isolated and purified from the seeds of Momordica charantia by following procedures:extraction with NaCl solution,ammonium sulfate fractionation,chromatography on SP-Sepharose Fast Flow,chromatography on Sephacryl S-100,and chromatography on Macro-Cap-SP.The results of SDS-PAGE showed that the two RIPs were a homogeneous single band,and molecular mass was estimated to be about 28 kDa.Double immunodiffusion showed that the two proteins had no cross reaction.
