摘 要:目的:检测肌球蛋白调节性轻链(MYL2)在慢性心衰大鼠心室肌的表达变化。方法:建立腹主动脉缩窄大鼠慢性心衰模型,采用免疫组化染色和集成光学密度(IOD)定量观测MYL2在心室肌的表达及其表达程度,并与假手术组对照分析。结果:心衰组大鼠左室收缩末压(LVSP)、左室内压上升最大速率(+dp/dtmax)、左室内压下降最大速率(-dp/dtmax)显著低于假手术组,左室舒张末压(LVEDP)显著高于假手术组(P均<0.01)。免疫组化染色示心衰组心室肌MYL2表达显著低于假手术组;心衰组染色标本的平均IOD为(47.12±12.08)×106,假手术组为(97.77±36.21)×106,前者显著低于后者(P<0.01)。结论:心室肌MYL2表达的显著降低可能是慢性心衰发生的重要机制。Objective:To detect the change of expression level of myosin regulatory light chain(MYL2) in chronic heart failure(CHF).Method:Rat model of CHF of abdominal aortic banding was established.Immunohistochemistry stain was carried on rat ventricular myocytes sample.Integrated optical density(IOD) was detected to determine the expression of MYL2.Results:The left ventricular end-systolic pressure(LVSP),the maximum increase rate of left ventricular pressure(+dp/dtmax),the maximum decrease rate of left ventricular pressure(-dp/dtmax) of CHF group were significant lower than those of pseudo-surgery group(P<0.01),the left ventricular end-diastolic pressure(LVEDP) of CHF group was significant higher than that of pseudo-surgery group(P<0.01).These data showed that the model was established successfully.In CHF group,the MYL2 protein level decreased significantly compared with pseudo-surgery group \[IOD:(47.12±12.08)×106 vs(97.77±36.21)×106,P<0.01\].Conclusion:MYL2 was down-expressed in CHF cardiocytes,and these finding suggested that MYL2 may play an important role in the development of CHF.
