普罗布考对糖尿病心肌病患者血清8-羟基脱氧鸟苷酸水平及心功能的影响

摘　　要：目的:探讨普罗布考抗氧化应激作用对糖尿病心肌病(DC)患者8-羟基脱氧鸟苷酸(8-OHdG)水平和心功能的影响。方法:(1)97例DC患者随机分为治疗组(61例)和对照组(36例),两组均在常规饮食控制基础上给予阿司匹林(100mg/天)、消心痛(10mg,3次/天)、缬沙坦(80～160mg/天)口服及胰岛素皮下注射,将血糖控制在在理想水平;治疗组加服普罗布考(0.5g,2次/天),疗程6个月。(2)两组治疗前和治疗6个月后采用ELISA测定其血清8-OHdG水平,并与40例健康体检者(正常组)进行比较。(3)采用超声心动图评价DC患者治疗前后心脏结构和功能变化。结果:(1)DC患者8-OHdG水平明显高于正常组(P<0.05);(2)治疗后,治疗组8-OHdG水平较治疗前明显下降(P<0.05),而对照组变化不明显(P>0.05);(3)治疗后,治疗组患者心脏收缩功能和舒张功能较治疗前明显改善,自身比较及与对照组比较均具有统计学意义(P<0.05)。结论:DC发病与氧化应激有关,抗氧化应激治疗对DC具有重要临床作用。Objective：To investigate the effects of Probucol in treatment of the cardiac function and the 8-OHdG in diabetic cardiomyopathy.Method：The Diabetic cardiomyopathy patients of（n=97）were divided in to the control group（36 patients）and the treated group（61 patients）randomly.Patients were given the following drugs repectively.Valsartan 80mg^160 mg.po,qd;Aspilin100mg.po,qd;Isosorbide dinitrate 10mg.po,tid;Probucol 0.5g po,bid was used in the treatment group for 6 months.with echocardiography was used to comparw some cardiac parameter before and after treatment the 8-OHdG by ELISA.Results：The findings showed higher elevated serum 8-OHdG in patients with diabetic myocardial disease.Diabetic cardiomyopathy group of the 8-OHdG was higher than those healthy blood donors（P0.05）.The results showed that the indites of left vertricular systole（LVEDV,LVESV,EF）and of diastolic function（E,A,E/A）had been significantly improved in the treated gruop.There was not significantly improved in the control group in the therapy group which was higher than that of control group（P0.05）.Conclusion：The findings suggest that these elevated serum 8-OHdG may play important roles in the pathogenesis of diabetic cardiomyopathy.Probucol was shown to be effective and safe in the treatment of diabetic cardiomyopathy.
