摘  要:目的:研究蒙药章古-3汤对腺嘌呤诱导大鼠CRF的干预作用及其机制。方法:60只Wister大鼠随机分为6组:空白对照组、病理模型组、章古-3汤高剂量治疗组(30g/kg.d)、章古-3汤中剂量治疗组(20g/kg.d)、章古-3汤低剂量治疗组(10g/kg.d)、尿毒清对照组,每组10只。以腺嘌呤200mg/kg.d灌胃建立CRF模型,造模同时各治疗组灌胃给予章古-3汤,尿毒清组给予尿毒清溶液10g/kg.d。连续灌胃24d后,检测生化指标(BUN、Scr、Na+、K+、Ca2+、P5+);观察肾组织病理改变;用免疫组化方法检测肾组织中TGF-β1的表达和分布。结果:经章古-3汤治疗的各组大鼠肾功能与同期病理模型组比较,有明显的改善(P<0.05),与尿毒清对照组相比无显著性差异。治疗组TGF-β1水平显著低于同期病理模型组(P<0.05),但皆高于正常对照组(P<0.01)。结论:蒙药章古-3汤对腺嘌呤导致CRF大鼠模型的肾功能有一定保护作用,其作用机制与抑制TGF-β1表达有关。Objective:To study the therapeutic effects of Traditional Mongolian Medicine Zhanggu-Ⅲ on adenine-induced chronic renal failure(CRF) in rat and explore the mechanism of Zhanggu-Ⅲ in preventing and treating CRF.Methods: Sixty wister rats were randomly divived into six groups:normal group,model group,Zhanggu-Ⅲ high dosage group(ZG-ⅢH),Zhanggu-Ⅲ middle dosage group(ZG-ⅢM),Zhanggu-Ⅲ low dosage group(ZG-ⅢL),and Niaoduqing group.each group had 10 rats.Each rat in model group,ZG-Ⅲ groups and niaoduqing group was infused with Adenine(20g/Kg·d) into stomach.in the same time,each rat in ZG-Ⅲgroup was also infused with high(30g/kg·d),middle(20g/kg·d),low(10g/kg·d) dosage forms ZG-Ⅲ into stomach for 24 days;the Niaoduqing group was given Niaoduqing liquid.The content of blood urea nitrogen(BUN),serum creatinine(Scr),biochemical indexes(Ca^2+,K^+,Na^+,P^5+),and changes in histopathology were observed after 24 days.Transforming growth factor-β1(TGF-β1) expression was examined by immuohistochemistry.Results: in ZG-Ⅲ treated group,the renal function is obviously lower than that in model group(P0.05).ZG-Ⅲ increased the level of Ca2+,reduced the levels of P5^+and K^+(P0.01),TGF-β1expression level is lower compared with model group.Conclusion:Traditional Mongolian Midicine ZG-Ⅲ can protect renal function of adenine-induced CRF rats.the mechanism may be relative to suppressing the expression of TGF-β1.
