摘  要:目的:研究灵杞黄斑颗粒对大鼠光损伤模型视网膜细胞凋亡相关基因c-fos、caspase-3及bcl-xL表达的影响。方法:SD大鼠随机分为正常对照组、光损伤给水组、光损伤给药组。采用白色荧光灯持续照射5h建立光损伤模型,于光照2·5h、5h及光照后1d及3d,通过逆转录-聚合酶链反应(RT-PCR)和免疫蛋白印迹(western blot)分析视网膜c-fos、caspase-3及bcl-xL的转录表达。结果:c-fos、caspase-3mRNA和蛋白表达下调,bcl-xL mRNA和蛋白表达上调。结论:灵杞黄斑颗粒可调控光损伤大鼠视网膜细胞凋亡相关基因的表达。Objective: To investigate the effect of Lingqi Huangban Granule (LQHB) on apoptosis-associated c-los, caspase-3and bcl-xL expression in light-induced retinal damage of rat. Methods: Sprague-Dawley (SD) rats were randomly divided into three groups: normal control group, light damage with vehicle group and light damage with LQHB group, whose retinal light impairment was induced by exposure to white fluorescent constant light for 5 hours. At 2-5h, 5h, after ld and 3d light exposure, the mRNA and protein expression of c-fos, caspase-3, bcl-xL were observed by RT-PCR and western bolt methods. Results : The mRNA and protein expression of c-fos and caspase-3was down regulated. The mRNA and protein expression of bcl-xL was up regulated. Conclusions : LQHB could regulate the expression of apoptosis-associated gene in light-induced retinal damage of rat.
