摘  要:[目的]建立反相高效液相色谱法对12个不同产地的辽细辛中马兜铃酸A进行痕量检测并比较其含量大小。[方法]采用超声提取法对辽细辛进行提取,通过反相高效液相色谱法对其含量进行测定。色谱柱为ZorbaxSB-C18(4.6 mm×150 mm,5μm),流动相为乙腈-水溶液(50∶50,V/V),检测波长为316 nm,柱温为室温,流速为1 ml/min。[结果]除凤城产地细辛外,其余细辛未检出含有马兜铃酸A;凤城产地细辛的马兜铃酸A在2.04~20.4 ng/ml浓度范围内线性关系良好。[结论]该方法简便快速,线性关系、灵敏度和重现性良好,回收率高,可用于辽细辛痕量马兜铃酸A含量的检测,所测产地的辽细辛总体质量较好。[Objective] The aim was to determine and compare the aristolochic acid A in RADIX ET RHIZOMA ASART from 12 different production areas by establishing Reversed-Phase High Performance Liqnid Chromatography(RP-HPLC).[Method] Ultrasonic extraction was used to extract the RADIX ET RHIZOMA ASART;and its content was determined by RP-HPLC.The chromatographic conditions were as follows: ZorbaxSB-C18(4.6 mm ×150 mm,5 μm),acetonitrile-water solution(V/V,50∶ 50),detection wavelength at 316 nm,room temperature,and flow rate at 1 ml/min.[Result] Except the RADIX ET RHIZOMA ASART in Fengcheng,no aristolochic acid A was detected.The conent of aristolochic acid A in RADIX ET RHIZOMA ASART produced in Fengcheng showed good linear relationship within the range of 2.04-20.4 ng/ml.[Conclusion] This method was rapid and simple with high recovery rate and good Linear relationship,sensitivity and repeatability.Thus,it could be used for the determination of aristolochic acid A content in RADIX ET RHIZOMA ASART.And the RADIX ET RHIZOMA ASART used in this research had relatively high quality.
