落地生根黄酮类成分分析及抗氧化活性研究

摘　　要：[目的]建立落地生根提取物中黄酮类成分的定性定量测定方法,并初步评价其体外抗氧化活性。[方法]将落地生根药材经水解后提取总黄酮,并采用紫外分光光度法(UV)测定总黄酮的含量、薄层色谱法(TLC)进行槲皮素和山柰酚的定性分析、Fenton反应法评价提取物的抗氧化活性、高效液相色谱法(HPLC)检测槲皮素及山柰酚的含量。[结果]UV法测得芦丁在11.0～110.0μg/ml浓度范围内线性关系良好(r=0.999 8),平均加样回收率为99.3%(n=6,RSD为1.3%);TLC可清晰检测出懈皮素与山柰酚;HPLC测得槲皮素在37.5～1 200.0 ng/ml浓度范围内线性关系良好(r=0.999 7)、山柰酚在62.5～2 000.0 ng/ml(r=0.999 4)浓度范围内线性关系良好;平均加样回收率分别为101.67%(n=6,RSD为1.92%)、98.68%(n=6,RSD为1.40%);结果显示提取物具有较大抗氧化能力,并成量效关系。[结论]该方法准确可靠、重复性好,可作为落地生根黄酮类成分定性定量的测定方法和落地生根药材的质量控制方法。[Objective] The aim was to establish a qualitative and quantitative analysis method for the flavonoids in HERB OF AIR-PLANT and its antioxidant activity.[Method] After hydrolysis,the total flavonoids were extracted and determined by UV spectrophotometry.TLC was applied to detect quercetin and kaempferol.Fention method was applied to determine the antioxidant activity.And the total quercetin and kaempferol content in the flavonoids was determined by HPLC.[Result] Rutin showed good linear relationship in the range of 11.0-110.0 μg/ml（r=0.999 8） with the average recovery of 99.3%（n=6,RSD=1.3%） by the UV spectrophotometry.Quercetin and kaempferol were detected by TLC.Quercetin showed good linear relationship in the range of 37.5-1 200.0 ng/ml（r=0.999 7） with the average recovery of 101.67%（n=6,RSD=1.92%）;and kaempferol showed good linear relationship in the range of 62.5-2 000.0 ng/ml（r=0.999 4） with the average recovery of 98.68%（n=6,RSD=1.40%）.The total flavonoids extracted from HERB OF AIR-PLANT showed high antioxidant activity,and antioxidant effect of total flavonoids from HERB OF AIR-PLANT increased as the concentration of total flavonoids enhanced.[Conclusion] This method was accurate and reliable,and could be used for the qualitative and quantitative determination of the flavonoids in HERB OF AIR-PLANT.
