摘 要:背景大量研究证实β1受体阻滞剂治疗急性冠状动脉综合征(ACS)有益,但作用机制尚不明确。目的探讨β1肾上腺素受体阻滞剂对健康人及ACS患者外周血单个核细胞一些炎症因子表达的影响。方法应用不同剂量的β1肾上腺素能受体阻滞剂艾司洛尔体外分别和健康人及ACS患者外周血孵化后,采用流式细胞术测定CD14+单个核细胞Toll样受体4(TLR4)的表达,RT-PCR法测定单个核细胞TLR4mRNA的表达,ELISA法测定血清中白细胞介素6(IL-6)、白细胞介素18(IL-18)水平。结果艾司洛尔下调了健康人及ACS患者外周血单个核细胞TLR4蛋白及mRNA、IL-6、IL-18的表达,并呈现一定的剂量依赖性。结论β1受体阻滞剂应用于健康人及ACS患者可抑制TLR4及其下游炎症因子的表达。Background The underlying mechanism of the effects of β1-adrenergic receptor blocker on the patients with acute coronary syndrome{ ACS) remain unclear. Objective To study the effect of β1-adrenergic receptor bloc- ker on inflammation cytokines expression in peripheral blood mononuclear ceils in patients with ACS and healthy vol unteers. Methods Peripheral blood samples of healthy participants and patients with ACS were administrated with different doses of esmolol. Toll-like receptor 4 (TLR4) protein and mRNA levels were respectively measured by flow cytometry and reverse transcription-polymerase chain reaction (RT-PCR). The level of interleukin (IL) 6, IL-18 were measured by ELISA. Results Both in patients with ACS and in health participants, the expressions of TLR4 protein, TLR4 mRNA, IL-6 and IL-18 in peripheral blood mononuclear cells were significantly depressed by esmolol in a dose-dependent manner. Conclusion The beneficail effects of β1-adrenergic receptor blocker on the pa-tients with ACS may be due to its inhibiting effect on the expression of TLR4 and inflammation cytokines.
