摘  要:目的建立测定莫西沙星人体血药浓度的反相高效液相色谱法,为临床合理用药提供参考。方法色谱柱为Diamonsil ODS柱(150 mm×4.6 mm,5μm),流动相为乙腈-水(含0.04 mol/L磷酸和0.4%三乙胺),流速为1.0 mL/min,柱温为25℃,检测波长为290 nm,灵敏度为0.01 AUFs。沉淀剂选用10%三氯醋酸,以峰面积外标法定量。结果莫西沙星的保留时间为8.7 min,质量浓度线性范围为0.05~5.00 mg/L,最低定量限为0.04 mg/L;平均回收率为96.22%~100.56%,绝对回收率为82.21%~93.65%;日内RSD为5.21%~6.30%,日间RSD为6.78%~7.56%。结论该方法简单、准确、快速和灵敏度高,可用于莫西沙星的人体血药浓度测定。Objective To establish a RP-HPLC method for the determination of moxifloxacin concentration in human plasma to provide reference for clinical rational drug use.Methods The separation of sample was performed on the Diamonsil ODS C18 column(150 mm×4.6 mm,5 um) with the mobile phase consisting of acetonitrile-water(containing 0.04 mol/L H3PO4 and 0.4%triethylamine).The flow rate was 1.0 mL/min,the column temperature was 25 ℃and the detection wavelength was 290 nm.The sensitivity was 0.01 AUFs.10%trichloroacetic acid was chosen as precipitator and the external standard method was used.Results The retention time of moxifloxacin was about 8.7 min.The calibration curve was linear within the concentration range of 0.05-5.00 mg/L.The lowest limit of quantitation was 0.04 mg/L.The average recovery rate was 96.22%-100.56%and the absolute recovery rate was 82.21%-93.65%.RSD for within day precision and between day precision were 5.21%-6.30%and 6.78%-7.56%.Conclusion The method is simple,accurate,rapid and highly sensitive,which could be used for the concentration determination of moxifloxacin in human plasma.
