摘 要:目的建立高效液相色谱法同时测定排石利胆颗粒中绿原酸和龙胆苦苷的含量。方法色谱柱:DiamonsilC18柱(4.6 mm×250 mm,5μm);流动相:甲醇-1%醋酸溶液(21∶79);流速:0.9 mL.min-1;柱温:25℃;检测波长:280nm。结果在32.512~325.12μg的范围内,绿原酸的进样量与峰面积呈良好的线性关系,其回归方程为Y=1 546.2X-1 904,r=0.999 9,平均回收率为100.314%,RSD=0.5%(n=6);在98.496~984.86μg的范围内,龙胆苦苷的进样量与峰面积呈良好的线性关系,其回归方程为Y=1 413.7X-9 226.2,r=1.000,平均回收率为99.527%,RSD=0.6%(n=6)。结论本方法操作简单,准确、专属性强、重现性好,可作为制订排石利胆颗粒质量标准的依据。Objective To establish an HPLC method for the determination of chlorogenic acid and gentioroside in Paishilidan Granules. Methods Diamonsil C18 column (4.6 mm x 250 mm,5 p.m) was adopted. The mobile phase was methanol - 0.1% acetic acid solution( 21:79 ). The flow rate was 0.9 mL · min -1. The column temperature was 25 ℃ and the detection wavelength was 280nm. Results The linear range of chlorogenic acid was 32.512- 325.12 μg and the regression equation was Y = 1 546. 2X - 1 904, r = 0. 999 9. The average recovery was 100.314% with the RSD = 0. 5 % ( n = 6 ) ; The linear range of gentioroside was 98. 496 - 984.86 μg and the regression equation was Y = 1 413.7X - 9 226. 2, r = 1. 000. The average recovery was 99. 527% with the RSD = 0. 6 % ( n = 6 ). Conclusion The method was simple, accurate, specificity, and can be used for the quality stand- ard of Paishilidan Granules.
