摘 要:目的观察赤芍总苷(TPG)对D-半乳糖(D-gal)诱导学习记忆功能低下衰老大鼠模型学习记忆能力作用,并探讨与衰老相关的氧化应激-自由基损伤、蛋白非酶糖基化反应及醛糖还原酶(AR)活性等作用。方法采用SD雌性大鼠,腹腔注射D-gal 500mg/kg,每天1次连续70天,诱导衰老大鼠模型;灌胃TGP 300mg/kg、150mg/kg和75 mg/kg,每天1次连续30天。采用Morris水迷宫测定法检测学习记忆能力,检测血浆果糖胺(FRA)、糖化血红蛋白(HbA1c)和晚期糖基化终产物(AGEs)含量及红细胞AR活性,以及大鼠脑组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)和AGEs含量。结果 D-gal致衰老大鼠学习记忆能力下降,红细胞AR活性增强,血浆FRA、HbA1c和AGEs含量明显增高,脑组织中SOD活性下降,MDA和AGEs含量明显增高,与正常对照组比较,差异均有统计学意义(P<0.01)。TGP 3个剂量能不同程度地改善衰老大鼠的脑功能及体内的病理生理改变(P<0.01或P<0.05)。结论 TGP通过抑制糖基化-氧化应激反应,降低早期和AGEs及脂质过氧化产物浓度;抑制AR活性,改善D-gal诱导衰老大鼠学习记忆能力。TGP的上述作用呈量效关系。Objective To investigate total paeony glycoside(TPG) on brain power of learning and memory and explore the oxidative stress-free radical damage,protein non-enzymatic glycation and aldose reductase activity and so on,in aging rats D-galactose-induced learning and memory dysfunction.Methods Sprague-Dawley female rats were induced aging intraperitoneal injected by D-galactose for 70 days,which 3 dose groups of TPG 300mg/kg,150mg/kg,75mg/kg intragastric administration for 30days were treated.The power of learning and memory was measured by Morris water maze,contents of fructosamine(FRA),glycosylated hemoglobin(HbA1c),advanced glycation end products(AGEs)in plasma and activity of aldose reductase(AR) in erythrocyte,and activity of superoxide dismutase(SOD),concentrations of malonaldehyde(MDA)and AGEs in brain tissue were determinated.Results Compared with the blank control rats,the power of learning and memory was decreased,activity of AR and levels of FRA,HbA1c and AGEs were increased in erythrocyte or plasma,activity of SOD was attenuated,concentrations of MDA and AGEs in brain tissue were increased;the above-mentioned differences were statistically significant(P〈0.01 or P〈0.05).The brain power of learning and memory were ameliorated and pathophysiological changes in vivo were improved by TPG(P〈0.01 or P〈0.05).Conclusion TPG can inhibit glycation-oxidative stress,reduce levels of early glycation products and/or AGEs,lipid peroxidation products,and repress activity of AR,ameliorate brain power of learning and memory in D-galactose-induced aging rats.The effects of TPG show dose-effect relationships.
