不同离心方法去除精浆对内蒙古白绒山羊精液冷冻保存效果的研究

摘　　要：山羊精液不同于其他家畜精液,它在含有卵黄的冷冻稀释液中的承受能力很低,山羊精浆里的酶能与卵黄发生反应,从而对精子产生毒副作用,所以去除精浆在山羊精液的冷冻保存中非常重要。为检测不同离心方法去除精浆对内蒙古白绒山羊精液冷冻保存效果,采用阿尔巴斯白绒山羊精液,用3种不同离心方法洗涤除去精浆后,对精液进行冷冻处理,结果表明:750r/min1次和2次离心处理组的精子冷冻解冻后的活率、质膜完整率、线粒体活性均显著高于1500r/min1次离心处理及对照组,而且750r/min1次离心处理组的精子顶体完整率也显著高于这2组;而1500r/min1次离心组的活率、顶体完整率、线粒体活性均与对照组无显著差异,但显著低于750r/min1次离心处理组,说明用不同离心方法去除精浆可以影响内蒙古白绒山羊精液冷冻保存效果。Goat semen is different from that of other domestic species in its limited tolerance to the inclusion of egg yolk in the freezing medium,and this tolerance depends on the presence of enzymes in the seminal plasma that react with egg yolk,producing toxic compounds to the spermatozoa.Therefore,removal of seminal plasma is very important in frozening of goat semen.In order to detect the effect of removal of seminal plasma with different centrifugation speed on cryopreservating of Inner Mongolia white cashmere goat semen.Goat semen was removed seminal plasma by three different centrifugation.The results from the sperms being removed seminal plasma by centrifugation at 750 r/min once or twice indicated that the rate of plasma membrane integrity and mitochondrial activity was apperently higher than the control group （P 0.05） and centrifugation at 1500 r/min once.Acrosome intact integrity rates of cryopreservated sperm in 750 r/min once group was apperently higher than that of 750 r/min twice group.The percentage of frozen-thawed sperm of the motility,acrosome integrity of the rates,mitochondrial activity of seminal plasma being removed by centrifugation at 1500 r/min once was not signifieant with the control group,while that was obviously lower than （P 0.05） by centrifugation at 750 r/min once group.The results showed that removal of seminal plasma by different centrifugation had an impact on cryopreservating of Inner Mongolian white cashmere goat semen.
