摘  要:目的观察色素上皮衍生因子(PEDF)对糖尿病大鼠视网膜谷氨酸(Glu)及γ-氨基丁酸(GABA)的影响,探讨PEDF视网膜神经保护作用的机制。方法链脲佐菌素(STZ)一次性腹腔注射建立糖尿病模型,随机分为正常对照组、糖尿病组、糖尿病PEDF干预组和干预对照组。PEDF干预组于成模后第4周玻璃体腔注射重组大鼠PEDF 2μL,干预对照组予等量且pH值相等的磷酸盐缓冲液,注射后48h采用柱前衍生高效液相色谱法检测大鼠视网膜Glu和GABA的含量,并分析其相关性。结果糖尿病PEDF干预组视网膜中Glu和GABA含量及Glu/GABA较糖尿病组和干预对照组明显降低,组间差别有统计学意义(P<0.05),PEDF干预组视网膜中Glu和GABA的含量虽然高于正常对照组,但Glu/GABA与正常对照组间差别无统计学意义。结论 PEDF可以降低糖尿病早期视网膜中兴奋性氨基酸堆积,维持Glu/GABA的平衡,从而对视网膜神经组织起到一定的保护作用。Objective To explore the effect of pigment epithelium-derived factor(PEDF) on retinal nerve tissue by investigating the changes of glutamate and γ-aminobutyric acid in the retina of diabetic rats.Methods SPF Sprague Dawley rats were divided into normal control group and diabetic group.Diabetic rat model was induced by intraperitoneal STZ administration.4 weeks later,10 diabetic rats were intravitreously injected with recombinant rat PEDF,another 10 diabetic rats were injected with an equal amount of phosphate-buffered saline(PBS) and served as control group.Animals were sacrificed and eye balls were collected 48 hours after injection.The levels of glutamate(Glu) and γ-aminobutyric acid(GABA) in the retina were quantitatively detected using high pressure liquid chromatography(HPLC).Result The levels of Glu,GABA and their concentration ratio in the retina of diabetic rats without PEDF injection were higher than that of the normal control rats and diabetic rats with intravitreous injection of PEDF.The differences were statistically significant(P0.05).The levels of Glu,GABA in the retina of diabetic rats with intravitreous injection of PEDF were statistically significantly higher than that of the normal control rats,but their concentration ratio of Glu/GABA was not statistically significantly different.Conclusion Intravitreal administration of PEDF may restrain the glutamate toxic effect on the retina of diabetic rats.PEDF has the protective effect on the retinal nerve tissue.
