榄香烯预防胃癌HGC-27裸鼠腹腔转移的研究

摘　　要：目的:研究榄香烯对胃癌HGC-27裸鼠腹膜种植转移的预防作用。方法:建立人胃癌HGC-27腹膜转移裸鼠模型,分别给与榄香烯、顺铂(DDP)及两者联合进行干预后,检测腹膜转移瘤腹膜肿瘤指数,并用免疫组织化学法检测转移瘤组织中增殖细胞核抗原Ki67、抗凋亡蛋白bcl-2的蛋白表达情况。结果:与对照组比较,榄香烯、DDP及榄香烯联合DDP干预组的腹膜转移瘤PCI指数均显著降低,其中以联合治疗组PCI指数降低最为显著(PCI指数为33,P<0.01)。同时,上述3种干预组转移瘤组织中Ki67、bcl-2蛋白阳性表达率显著降低,其中以联合治疗组Ki67、bcl-2蛋白阳性表达率降低最为显著(57.14%和34.28%,均P<0.05)。结论:榄香烯单独或者联合DDP应用对于人胃癌HGC-27裸鼠腹腔转移有显著的预防作用。Objective：To study the preventive effect of Elemene on HGC-27 gastric cancer peritoneal metastasis in nude mice and provide the experimental basis for expanding its novel clinical implications. Methods： The HGC-27 human gastric cancer nude mouse model of peritoneal metastasis was established, and administrated with Elemene, cisplatin（DDP） or Elemene combined with DDP. The peritoneal cancer index of peritoneal metastases was determined, and immunohistochemistry was performed to detect the expression of Ki67, bcl-2 protein expression. Results： HGC-27 human gastric cancer nude mouse model of peritoneal metastasis after elemene and DDP given after the intervention, PCI index of peritoneal metastases were significantly different, which combined treatment group, Bcl-2 protein expression differences to some extent. Conclusions： Elemene in human gastric cancer HGC-27 for peritoneal metastasis in nude mice have a clear preventive effect.
