摘 要:目的:探讨不同神经发育阶段铅暴露对海马神经元数目的影响。方法:将Wistar大鼠随机分为孕期染铅组、断乳后染铅组和对照组,处理组经水给予PbCl2400μmol/L,对照组自来水。至观察终点采用免疫组化法检测海马CA3区神经元数目。结果:仔鼠出生第1天,孕期染铅组与对照组神经元数目均数分别是315.52±42.12、435.25±25.38,明显低于对照组(P<0.05);至2月龄,孕期染铅组仔鼠神经元均数仍显著低于对照组(P<0.05),而出生断乳后染铅组神经元均数与对照组相比,差别无统计学意义P﹥0.05,其均数分别是45.00±7.83、58.25±7.85和58.74±4.92。结论:孕期铅暴露可导致仔鼠海马神经元数目减少,而出生后铅暴露则对海马组织神经元数目没有明显影响。Objective:To discuss the effect of lead exposure on neuron numbers of hippocampus at different neural developmental stages of rats.Methods: The Wistar rats were divided into lead exposure group during pregnancy,lead exposure group after weaning and control group.The lead exposure groups were supplied with water containing 400 μmol/L of lead acetate and the control group was supplied with drinking water from the animal house.At the end point of observation,the immunohistochemical method was used to detect the hippocampus′s neuron number of the CA3 region.Results: The means of the neuron numbers of the 1-day-old rats in lead exposure group during pregnancy and the control group were 315.52±42.12,435.25±25.38 respectively,the number of treatment group was smaller than that of control group and the difference was statistically significant,P0.05;the means of the neuron numbers of two-month-old rats in lead group during pregnancy,lead group after weaning and the control group were 45.00±7.83,58.25±7.85 and 58.74±4.92 respectively.Compared with the control group respectively,the former till significantly reduced,P0.05 and the latter had no obvious changes,P0.05.Conclusion: Lead exposure during pregnancy can result in reduction of neuron cell number of hippocampus of rats,while lead exposure after weaning has no significant effect on neuron number of the hippocampus.
