摘 要:目的:运用脉冲场凝胶电泳(PFGE)技术对湖北2008年分离的沙门菌菌株进行分析,探讨湖北沙门菌的分布、流行及变异情况。方法:对分离的沙门菌利用全自动生化分析系统、血清分型、及脉冲场凝胶电泳(PFGE)分子生物学技术进行研究。结果:18份测试株用XbaI酶切后产生3个带型。结论:通过脉冲场凝胶电泳(PFGE)全基因DNA指纹图谱分析,可追溯其同源性。通过18株伤寒菌株的PFGE分型图谱分析,其中相似系数100%15份,占83.33.%,可确认为相同病原菌感染。由此说明PFGE是一种灵敏、可靠的研究沙门菌分子流行病学基因分型方法。Objective:To explore the distribution,popularity and variation of salmonella in Hubei through analyzing the salmonella strains isolated in 2008 by pulsed field gel electrophoresis(PFGE).Methods: Biochemical full autoimmunization analyzing system,typing of blood serum,typing of drug sensitivity and PFGE molecular biology technology were used to make a research on separated salmonella.Results: 18 tested strains produced 3 banding patterns after being divided by enzyme XbaI.Conclusion: The homology of salmonella of Hubei in 2008 can be traced by analyzing the whole genome DNA fingerprint by PFGE.Through analysis of PFGE typing map among the18 typhoid strains,15 strains(83.33%) are found complete homology,which confirms that they are infected by the same pathogenic bacteria.So PFGE is a sensitive and reliable method to study salmonella genotyping in molecular epidemiology.
