摘 要:目的:探讨脂蛋白酯酶(LPL)在妊娠期糖尿病孕妇脂肪及胎盘中的表达及其异常表达的意义。方法:采用定量RT-PCR及Western bolt方法检测妊娠期糖尿病妇女(GDM组)和正常妊娠妇女(对照组)剖宫产前的血脂水平及其皮下脂肪组织及胎盘组织的LPL蛋白表达情况。结果:GDM组血甘油三酯含量为(3.29±1.36)mmol/L,对照组为(2.34±1.35)mmol/L,两组比较P<0.05。LPL mRNA在GDM组脂肪组织中的相对表达水平为4.9×10-2±1.1×10-2,与对照组的5.5×10-2±1.3×10-2比较差异无显著性;LPL mRNA在GDM组胎盘组织中的相对表达为3.2×10-2±0.7×10-2,对照组为1.0×10-2±0.5×10-2,差异有统计学意义(P<0.05)。Western blot显示,两组孕妇脂肪组织中LPL表达无明显差异,但GDM患者胎盘组织中LPL表达较对照组明显增强。结论:妊娠期糖尿病孕妇胎盘中LPL表达增强可能是引起母体到胎儿的脂质转运异常的因素之一。Objective:To explore the expressions of lipoprotein lipase(LPL) in placental tissue and adipose tissue of pregnant women with gestational diabetes mellitus(GDM) and the significance of its abnormal expression.Methods:Quantitative RT-PCR and Western-blot were used to detect the blood lipids levels and expression levels of LPL in subcutaneous adipose tissue and placental tissue in pregnant women with GDM(GDM group) and normal pregnant women(control group) before cesarean section.Results:The levels of triglyceride in GDM group and control group were(3.29±1.36) mmol/L and(2.34±1.35) mmol/L,respectively,there was significant difference between the two groups(P0.05).The relative expression levels of LPL mRNA in adipose tissues of GDM group and control group were(4.9×10-2±1.1×10-2) and(5.5×10-2±1.3×10-2),respectively,there was no significant difference between the two groups;the relative expression levels of LPL mRNA in placental tissues of GDM group and control group were(3.2×10-2±0.7×10-2) and(1.0×10-2±0.5×10-2),respectively,there was significant difference between the two groups(P0.05).Western blot showed that there was no significant difference in expression level of LPL in adipose tissue between GDM group and control group,but the expression level of LPL in placental tissue in GDM group was significantly higher than that in control group.Conclusion:The high expression of LPL in placental tissue of pregnant women with GDM may be one of the factors inducing maternal-fetal lipid transfer disorder.
