摘 要:目的:测定线粒体融合蛋白2(Mitofusin 2,Mfn2)在正常早孕妇女和不明原因胚胎停止发育患者绒毛组织中的表达差异,探讨Mfn2在胚胎发育过程中的可能作用。方法:采用HE染色观察正常早孕妇女(正常早孕组,30例)和不明原因胚胎停止发育患者(胚胎停育组,35例)绒毛组织形态;采用免疫组织化学技术(SABC法)检测Mfn2在两组妇女绒毛组织中的定位表达和表达强度;同时采用实时荧光定量RT-PCR(qRT-PCR)技术检测两组妇女绒毛组织中Mfn2的相对表达。结果:Mfn2在正常早孕组和胚胎停育组绒毛合体滋养细胞和细胞滋养细胞中均有阳性表达,在细胞滋养细胞中表达尤为明显,而在间质细胞中无明显表达,且Mfn2阳性表达主要分布在细胞浆中。胚胎停育组的绒毛组织中Mfn2的相对表达量(0.006 8±0.003 2)显著低于正常早孕组(0.023 9±0.005 5),差异有统计学意义(P<0.01)。结论:Mfn2在不明原因胚胎停止发育患者绒毛组织中的表达水平下降,提示Mfn2可能与胚胎发育相关,绒毛滋养层细胞中Mfn2的正常表达可能对维持早期妊娠起到一定作用。Objective:To detect the difference of expressions of Mitofusin 2(Mfn2) in villous tissue of normal early pregnant women and cases with missed abortion,explore the probable effect of Mfn2 on the course of embryonic development.Methods:HE staining method was used to observe the morphology of villous tissue in the normal early pregnant women(early pregnancy group,30 cases) and cases with missed abortion(missed abortion group,35 cases);immunohistochemical technique(SABC method) was used to detect the location and expression level of Mfn2 in villous tissues of the two groups;the relative expression of Mfn2 in villous tissues of the two groups were detected by qRT-PCR.Results:Mfn2 expressed positively in villous syncytiotrophoblast and cytotrophoblast of early pregnancy group and missed abortion group,especially in cytotrophoblast,but Mfn2 didn't express in interstitial cells,the positive expression of Mfn2 mainly distributed in cytoplasm.The relative expression level of Mfn2 in villous tissue of missed abortion group was(0.006 8±0.003 2),which was significantly lower than that of early pregnancy group(0.023 9±0.005 5)(P0.01).Conclusion:The expression level of Mfn2 in cases with unexplained missed abortion decreases,which indicates that Mfn2 may be related to embryonic development.The normal expression of Mfn2 in syncytiotrophoblast may play a certain role to maintain early pregnancy.
