摘 要:目的探讨脑源性神经营养因子(BDNF)基因val 66met多态性与2型糖尿病(T2DM)伴抑郁的关系。方法用PCR及聚丙烯酰胺凝胶电泳方法检测246例T2DM(DM组)患者和186例健康体检者(C组)的BDNF基因val 66met多态性。用汉密尔顿抑郁量表(HAMD)评估T2DM患者,HAMD≥17分者组成T2DM伴抑郁(D)组。结果与C组相比,DM和D组患者以及其中的男性患者的等位基因G(val)和基因型G/G分布频率增高,等位基因A(met)和基因型A/A降低(P<0.05)。而两组女性患者的基因型A/A和基因型G/G均显著低于C组(P<0.05)。C组中,男性等位基因A和基因型G/A分布频率显著高于女性,等位基因G和基因型G/G显著低于女性(P<0.05)。D组中,男性抑郁发病率显著低于女性(P<0.05)。结论 BDNF基因val 66met多态性与T2DM、T2DM伴抑郁可能存在相关性;女性T2DM患者较男性可能易伴发抑郁。Objective To study the association of polymorphism of brain-derived neurotrophic factor val 66 met and depression in patients with type 2 diabetes mellitus(T2DM). Methods Using PCR technique,val 66 met polymorphism of BDNF was determined in 246 T2DM(group DM) and 186 healthy controls(group C).HAMD seals of depression in patients with T2DM were evaluated,who were defined as depression cases(group D) with the HAMD seals≥17.Results Compared to group C,the frequencies of G and G/G were higher,but A allele and A/A genotype were lower,in groups of DM and D and the male cases of the two groups(P0.05).The frequencies of G/G and A/A were lower in the females of groups of DM and D than those in group C(P0.05).The frequencies of A and G/A were higher,but G and G/G were lower,in the males than those in females in group C(P0.05).The incidence of depression was higher in females than that in males in group D(P0.05). Conclusion The polymorphism of BDNF gene val 66 met is associated with T2DM and depression in T2DM patients.Male T2DM patients has less incidence of developing depression than T2DM females.
