摘  要:目的使用程序低氧舱建立慢性间歇低氧(chronic intermittent hypoxia,CIH)大鼠模型,超声心动图评价大鼠心脏结构和功能改变。方法 16只大鼠随机分为2组,每组8只,分别为CIH组和对照组。CIH组大鼠置于程序低氧舱内,按程序设计予以每天8h,共35天的间歇低氧,每一低氧复氧循环时间为1min。对照组置于同样的舱内予以空气脉冲供气。测定低氧舱内不同循环时间的氧浓度。实验后,分别对两组大鼠进行超声心动图检查。结果程序低氧舱内的氧浓度呈周期性波动于(6.8±0.1)%~(20.3±0.3)%之间,达到低氧事件60次/min的环境设置,与重度睡眠呼吸暂停低通气综合征相对应。CIH组大鼠存在左室射血分数减低[CIH组:(85.5±1.5)%,对照组:(91.5±0.8)%,P<0.05]和肺动脉流速增快[CIH组:(1.126±0.041)m/s,对照组:(0.903±0.038)m/s,P<0.05]等表现。结论程序低氧舱内的氧浓度变化能够模拟阻塞性睡眠呼吸暂停低通气综合征(obstructive sleep apnea hypopnea syndrome,OSAHS)的低氧复氧模式。大鼠CIH模型是较为理想的OSAHS动物模型,超声心动图适合用于监测CIH大鼠心脏结构和功能的改变。Objective To establish a rat chronic intermittent hypoxia(CIH) model and evaluate its cardiovascular changes by echocardiogram.Methods Sixteen male rats were randomly divided into two groups:CIH group and control group with 8 rats in each group.The rats in CIH group were placed in an animal chamber subjected to intermittent hypoxia for 35 days with 60 seconds per cycle,8 hours per day and 7 days per week.While the rats in control group were placed in the same chamber with pulse air.Oxygen concentrations in the chamber at different time points were monitored.After the experiment,echocardiogram was done to measure cardiac changes.Results Oxygen concentration in the chamber periodically fluctuated between(6.8±0.1)% to(20.3±0.3)%.It could mimic severe sleep apnea hypopnea syndrome with hypoxia events of 60 times/h.The value of left ventricular ejection fraction(LVEF) in CIH group was lower than that in control group [(85.5±1.5)% vs(91.5±0.8)%,P0.05].The value of pulmonary arterial flow(PAF) in CIH group was higher than that in control group [(1.126±0.041)m/s vs(0.903±0.038)m/s,P0.05].Conclusions The automatically controlled hypoxic chamber used for rat CIH model could mimic the hypoxia and re-oxygenation process in patients with obstructive sleep apnea hypopnea syndrome(OSAHS).The rat CIH model with cardiac dysfunction was ideal for the study on complications of OSAHS.Echocardiogram was suitable for monitoring cardiac structure and function changes of CIH rats.
