摘  要:目的探讨低强度脉冲超声(low-intensity pulsed ultrasound,LIPUS)促进骨髓间充质干细胞(bone mesenchymal stemcells,BMSCs)增殖的可行性及筛选实验的最佳参数。方法选用频率0.6 MH_z,强度为0.03、0.1、0.2、0.3 W/cm^2的LIPUS分别辐照体外培养的第三代兔骨髓间充质干细胞5、10、15、20、30 min,24 h后观察细胞生长情况,四甲基偶氮唑蓝(MTT)法和流式细胞仪检测LIPUS辐照后细胞增殖活性和细胞周期。结果各参数组经过LIPUS辐照后24 h观察细胞分裂增殖均较空白对照组明显活跃,细胞集落更为密集。细胞增殖活性和细胞增殖指数均明显高于空白对照组(P<0.05),其中以0.2 W/cm^2,10 min辐照后促进增殖效果最为显著。结论 LIPUS辐照一定时间后能够促进BMS(Cs增殖,不同的辐照强度和辐照时间促进其增殖的效应不一,其中强度为0.2W/cm^2,辐照10 min剂量促进增殖效果最佳。Objective To investigate the feasibility of promoting bone mesenchymal stem cells(BMSCs)proliferation by low intensity pulsed ultrasound(LIPUS)and screen out the most optimal parameter of LIPUS. Methods The third generation of BMSCs in vitro was respectively irradiated by LIPUS of different intensity of 0. 03,0.1,0. 2 and 0.3 W/cm2 , 0.6 MHz for 5,10,15,20 and 30 min. Growth of cells was observed under light microscope. Proliferation ac tivity was determined by MTT and cell cycle changes were analyzed using the flow cytometry after 24 h post-irradia- tion. Results Cell proliferation of different parameter groups was significantly more active than that of the control- group and more intensive cell colonies formed in LIPUS groups after 24 h post-irradiation. Both the cell proliferation activity and the cell proliferation index(PI) in different parameter groups were remarkably higher than that in the con trol group (P〈0.05) ; The effect of proliferation promotion posvirradiation with 0.2 W/cm2 for 10min is the most prominent. Conclusions LIPUS can promote proliferation of BMSCs after sometime post-irradiation. The promotion effect is different with intensity and irradiation time change of LIPUS. 0.2 W/cm2 for 10min is the most prominent parameter.
