HOXA10基因在子宫内膜异位症患者在位及异位子宫内膜组织中的表达及其意义

摘　　要：目的探讨HOXAl0基因在子宫内膜异位症（内异症）患者在位及异位内膜组织中的表达水平及其临床意义。方法选取2009年1月至2010年8月于佛山市妇幼保健院因内异症行腹腔镜手术治疗的患者30例，留取宫腔内膜组织（内异症在位内膜组）和异位内膜组织（内异症异位内膜组）；同期因卵巢囊肿及输卵管因素不孕行腹腔镜手术治疗患者30例为对照组，留取宫腔内正常内膜组织。分别采用实时荧光定量PCR技术、免疫组化法和蛋白印迹法检测各组内膜组织中HOXAl0mRNA及蛋白表达水平。结果内异症在位内膜组HOXAl0mRNA及蛋白表达水平分别为0．614-0．07和0．47±0．05，内异症异位内膜组分别为0．644-0．06和0．504-0．05，对照组分别为1．224-0．14和1．424-0．14；内异症在位和异位内膜HOXAl0mRNA及蛋白表达水平明显低于正常内膜中的表达水平，差异均有统计学意义（P均〈0．01）；内异症在位和异位内膜组的HOXAl0mRNA及蛋白表达水平比较，差异则无统计学意义（P〉0．05）。免疫组化法检测内异症在位和异位内膜组织的问质和腺细胞中HOXAl0表达均降低。结论HOXAl0基因在内异症在位及异位内膜组织中均呈低表达状态，可能与内异症的发病和不孕有关。Objectives To investigate the expression and clinical significance of HOXA10 gene in the eutopic and ectopic endometrium of endometriosis. Mehtods Between Jan. 2009 to Aug. 2010, 30 patients with endometriosis undergoing laparoscopic surgery in Maternal and Children＇s Hospital of Foshan. Eutopic and eetopic endometrium were obtained. In the mean time ,30 patients with benign ovary cyst or tubal infertility undergoing laparoscopic surgery were selected as controls. Their uterine endometrium were obtained real-time fluorescent quantitation, western blot and immunohistochemistry technique were used to detect mRNA and protein expression of HOXA10 gene in the eutopic endometrium group, ectopie endometrium group and control group. Results The mRNA and protein expression of HOXA10 gene were 0. 61 ~0. 07 and 0. 47 ~0. 05 in the eutopic endometrium of endometriosis,0. 64 ~0. 06 and 0. 50 ~0. 05 in eetopic endometrium of endometriosis, which were significantly lower than 1.22 ~ 0. 14 and 1.42 ＋- 0. 14 in control group （P 〈0. 01 ）. However, the mRNA and protein expression of HOXA 10 between eutopic and ectopie endometrium of endometriosis did not reach statistical difference （ P 〉 O. 05 ） . The expression of HOXA10 in eutopic and ectopic endometrium of endometriosis were decreased by immunohistochemistry staining. Conclusion The lower expression of HOXA10 gene in the eutopic and ectopie endometrium of endometriosis might be associated with pathogenesis and infertility of endometriosis.
