摘 要:目的:观察雷公藤提取物对大鼠心脏急性损伤的影响。方法:将SD大鼠随机分为空白对照组和雷公藤提取物低(137 mg/kg)、中(275 mg/kg)、高(550 mg/kg)剂量组,各组ig给药,分别于给药前(0 h)和给药后1、2、4、8、12、24h检测血钾、心电图及心功能指标,并采用HE染色法观察24h大鼠心肌组织病理变化。结果:与给药前或空白对照组比较,各剂量组均出现血钾降低、心电图指标:心率降低,Q-T间期、S-T间期、QRS间期延长和ST段下降;心功能指标:左室收缩压(LVSP)降低、左室内压最大上升/下降速度(±dp/dtmax)降低、左室舒张末压(LVEDP)升高。各指标均在4~8h变化最显著,且给药剂量越大,各指标变化出现时间越早,变化程度越明显。HE染色显示,给药后低剂量组大鼠心肌未见病理变化,中剂量组大鼠心肌有轻微损伤、高剂量组大鼠心肌有明显损伤。结论:雷公藤提取物在137~550 mg/kg剂量范围内灌胃给药后对大鼠心脏有急性损伤作用,且给药剂量越大,损伤越明显。血钾降低可能为心脏急性损伤的原因之一。Objective: To observe the effects of Tripterygium wilfordii extract on acute myocardial injury in rats.Methods: Sprague-Dawley rats were randomly divided into four experimental groups:control group,low(137mg/kg),middle(275 mg/kg),high(550 mg/kg) dosage groups.The rats were administrated by gastric perfusion.The level of serum potassium,the index of electrocardiogram(ECG) and heart function were separately tested at 0,1,2,4,8,12,24 h after administration.The pathology changes were evaluated by Haematoxylin-eosin(HE).Results: Each tested index experienced a change in low,middle,high dosage groups by comparing them with the control group.These changes included,for example,the level of serum potassium decreased,ECG index such as heart rate decreased,QT interval,ST interval,QRS interval prolonged,ST segment decreased,heart function index such as left ventricular systolic pressure(LVSP) decreased,maximum rates of increase and decrease of left ventricular pressure(±dp/dtmax) decreased and the left ventricular end diastolic pressure(LVEDP) increased.Each index was the most significantly changed at 4~8 h.Morever,if the dosage was more,the time of index change was earlier and the degree of change was deeper.In addition,it was not seen the rat myocardial tissue had pathological changes in low dosage group.The rat myocardial tissue had light injuries in middle dosage group,and had obvious injuries in high dosage group.Conclusion: The Tripterygium wilfordii extract could induce acute myocardial injury,and as the dosage was increased,the degree of injure was more obvious.The decreased serum potassium level may be one of reasons of acute myocardial injury.
