摘 要:目的:研究模拟微重力(SMG)对人牙髓干细胞(hDPSCs)在聚乳酸-羟基乙酸(PLGA)支架上黏附的影响。方法:采用酶消化法培养hDPSCs,并进行鉴定。将hDPSCs在模拟微重力条件接种于PLGA支架,接种细胞数分别为0.2×106、0.4×106、0.8×106、1.2×106个/支架,以常规条件接种作为对照组。接种24h后通过细胞计数检测细胞粘附率;扫描电镜(SEM)观察细胞形态。结果:模拟微重力条件接种组随接种细胞数的增多,黏附率显著升高(P<0.01),常规条件接种组黏附率略有降低(P<0.05);当接种细胞数为0.4×106、0.8×106、1.2×106/支架时,实验组高于对照组(P<0.05或P<0.01);为0.2×106/支架时,对照组高于实验组(P<0.01)。SEM显示模拟微重力条件接种组的细胞具有丰富的表面微绒毛。结论:模拟微重力条件下hDPSCs在PLGA支架上的黏附率高于常规条件。Objective: To investigate the adhesion efficiency of human dental pulp stem cells(hDPSCs) on PLGA scaffolds on two different conditions.Methods: hDPSCs were seeded on PLGA scaffolds with the same quantities,both on the conditions of conventional environment and simulated microgravity.After 24 hours,the cells-scaffold compositions were obtained.SEM was employed to observe the cells morphology and cells counting to determine the quantities of adherence cells.Then the adhesion efficiencies were calculated.Results: The adhesion efficiencies decreased with the increase of seeding cells on the conventional condition(P0.05),and significant increased on the simulated microgravity condition(P0.01).The adhesion efficiencies of simulated microgravity condition were higher than the conventional condition(P0.01).Cells seeded on the simulated microgravity displayed more abundant microvilli.Conclusion: The simulated microgravity would promote the adhesion of hDPSCs on PLGA scaffolds.
