摘 要:目的:研究精元康胶囊对骨髓抑制小鼠造血祖细胞增殖的影响的作用机制。方法:雄性昆明种小鼠70只,随机分为正常组、模型组、利可君片组、贞芪扶正胶囊组、精元康胶囊高、中、低剂量组等7组。用环磷酰胺(CTX)建立骨髓抑制的小鼠模型。前3 d每日上午ip CTX 100 mg.kg-1(正常组注射等量生理盐水),下午ig不同药物。3 d后,仅在下午ig不同药物。应用体内扩散盒植入的方法进行培养3系造血祖细胞,在奥林巴斯光镜下计数3系造血祖细胞集落数。结果:小鼠在注射CTX造模后,3系集落数与正常组相比均有明显降低(P<0.05);精元康胶囊各剂量组均能提升骨髓抑制小鼠3系祖细胞(粒单系造血祖细胞CFU-GM、红系造血祖细胞CFU-E/BFU-E、巨核系造血祖细胞CFU-Meg)集落产率(P<0.05),而且呈剂量依赖性,其中高剂量组对3系集落的提升作用最明显(P<0.05)。对CFU-E产率的影响,利可君片组、精元康胶囊中剂量组、贞芪扶正胶囊组均与精元康胶囊低剂量组有明显差异(P<0.05),但3组之间没有明显差异。对BFU-E的影响,精元康胶囊高剂量组优于其他药物组,其他各组间无明显差异;从CFU-GM的产率比较可知,利可君片组低于精元康胶囊高剂量组,但优于其他各组。精元康胶囊中剂量组和贞芪扶正胶囊组效果优于精元康胶囊低剂量组(P<0.05),但两者之间差异不明显。各药物组对CFU-Meg产率的影响,以精元康高剂量组集落产率最高,利可君片组次之,与其他各药物组有显著差异(P<0.05)。结论:精元康胶囊可增加3系造血祖细胞集落数,促进造血干细胞的增殖和分化。Objective:To study the effects of Jingyuankang capsule(JYK capsule) on the mouse hemopoiesis ancestor cells in cyclophosphamide(CTX) induced myleosuppression.Method:Group: Seventy male Kunming mice were randomly divided into normal group,model group,Likejun group,Tonic Capsule group,JYK capsule high dose group,JYK capsules medium dose group,JYK capsule low dose group.The mouse models of CTX-induced myleosuppression were adopted.All groups mice received an intraperitoneal injections of 100 mg·kg-1 CTX while normal group's mice received NS as control in the morning for 3 days,and intragastric infusion with different drugs in the afternoon.then,only done those as in the afternoon.Model: the progenitor cell colony,progenitor cells cultured by implantation in vivo diffusion chamber method and count in the light microscope Olympus.Result: The results of hematopoietic progenitor cells in vivo culture: JYK each dose group could improve bone marrow progenitor cells(granulocyte monocyte progenitor cell CFU-GM,erythroid progenitor cells CFU-E/BFU-E,megakaryocyte hematopoietic progenitor cell CFU-Meg) colony yields rate of mices.JYK capsule high dose group was more produced.Conclusion: JYK capsule can promote the proliferation of hematopoietic stem cells by humoral factor or some of bioactive materials to increase three progenitor cells colony and promote the proliferation of damaged blood cells.
