摘  要:目的:探讨川芎嗪对高脂高糖饮食与链脲佐菌素(STZ)诱致大鼠肾脏损伤保护作用的抗氧化机制。方法:SD大鼠70只。除正常组外,其余大鼠均给予高脂-高糖饲料喂养4周,再给予STZ(40 mg.kg-1,ip)造模,72 h后测定空腹血糖,将血糖>16.67 mmol.L-1的大鼠随机分为4组:模型组,二甲双胍组(250 mg.kg-1),川芎嗪低、高剂量组(80,160 mg.kg-1),ig,连续8周。末次给药后,收集24 h尿,测定尿量和尿蛋白量;测定血肌酐(SCr)、尿素氮(BUN)等变化;测定肾组织丙二醛(MDA)含量及超氧化物歧化酶(SOD)和Na+-K+-ATP酶活性;测定糖化血红蛋白(HbAlc)和糖基化产物(AGEs)的含量。结果:模型大鼠SCr,BUN含量明显升高(P<0.01);肾组织MDA含量升高,SOD活性降低,Na+-K+-ATP酶活性减弱(P<0.01);HbAlc和AGEs明显升高(P<0.01)。川芎嗪高、低剂量治疗8周后,明显降低SCr和BUN含量(P<0.05);减低肾组织MDA含量,提高SOD和Na+-K+-ATP酶活性(P<0.01,P<0.05);明显降低HbAlc和AGEs的含量(P<0.01)。结论:川芎嗪对高糖高脂饮食与STZ诱致大鼠肾损伤具有保护作用,其机制可能与增强肾脏组织抗氧化作用有关。Objective:To investigate the protective effects and its antioxidant mechanism of tetramethylpyrazine on renal injury induced by high-fat diet and streptozotocin(STZ).Method:Seventy rats were randomly divided into normal control group(14 rats) and model group.All model rats were fed by high-fat diet for 4 weeks and then injected with STZ(40 mg·kg-1,ip).Blood glucose levels were measured 72 h after STZ injection using a one-touch glucometer.Rats with fasting blood glucose level above 16.67 mmol·L-1 were randomly divided into four groups: model control,metformin(250 mg·kg-1)and tetramethylpyrazine(80,160 mg·kg-1) groups.All rats were treated for 8 weeks.At the end of experiment,urine was collected from the rats for 24 h to measure the urinary volume and protein.And then blood was collected to detect the changes of creatinine(Cr) and blood urea nitrogen(BUN),contents of glycated hemoglobin(HbAlc) and advanced glycation end products(AGEs).Contents of AGEs and malondialdehyde(MDA),activity of superoxide dismutase(SOD) and Na+-K+-ATP enzyme in kidney tissues were tested respectively.Result: Compared with normal control,the levels of SCr,BUN were significantly increased in the models(P0.01),content of MDA increased,while activity of SOD and Na+-K+-ATP enzyme decreased significantly(P0.01) in kidney tissues,and contents of HbAlc,concentrations of AGEs in serum and kidney were increased markedly.Treated with Tetramethylpyrazine(80,160 mg·kg-1) for 8 weeks,the levels of SCr and BUN in blood were decreased(P0.05);and the MDA content in kidney tissues was reduced,activities of SOD and Na+-K+-ATP enzyme were also enhanced respectively(P0.01,P0.05),and contents of HbAlc,concentrations of AGEs in serum and kidney were decreased markedly.Conclusion:Tetramethylpyrazine appears protective effect for renal injury rats induced by high-fat diet and STZ,and its mechanism may be concerned with increasing the antioxidative ability.
