摘  要:目的黄芪多糖有促进血管新生作用,通过探讨黄芪多糖胶原促进周围组织中血管新生与胶原代谢的关系及其对相关因子的调控作用,为该胶原作为促进血管新生与组织愈合的支架提供依据。方法以交联剂将黄芪多糖与空白胶原进行1∶1(W/W)共价结合,形成黄芪多糖胶原。取10周龄SD大鼠20只,雌雄各半,体重200~250 g。于脊柱两侧作纵形切口,皮下向两侧游离形成"口袋",左侧植入大小为5 mm×5 mm×5 mm的黄芪多糖胶原作为实验组,右侧植入相同大小空白胶原作为对照组。术后观察大鼠一般情况,于3、7、14、21 d分别处死5只大鼠,取胶原周围肉芽组织,组织学染色观察并计数毛细血管,测量羟脯氨酸(hydroxyproline,Hyp)含量,实时荧光定量RT-PCR测定血管生成素1(angiopoetin 1,Ang1)、基质金属蛋白酶9(matrix metalloproteinases 9,MMP-9)及基质金属蛋白酶抑制物1(tissueinhibitors of metalloproteinases 1,TIMP-1)mRNA的表达。结果大鼠均存活至实验完成。术后两组胶原周围肉芽组织中毛细血管逐渐增多,14 d时达峰值,实验组均多于对照组(P<0.05)。两组Hyp含量逐渐升高,各时间点实验组均高于对照组(P<0.05)。除21 d外,实验组术后3、7、14 d的Ang1及MMP-9 mRNA表达均显著高于对照组(P<0.05)。术后对照组TIMP-1 mRNA表达逐渐减弱,实验组呈逐渐增强趋势;除术后7 d外,两组比较差异均有统计学意义(P<0.05)。结论黄芪多糖胶原具有促进血管新生与胶原合成作用,机制之一是通过调节组织中Ang1、MMP-9及TIMP-1表达而实现。Objective Astragalus polysaccharide(APS) has promoting angiogenesis function.To explore the effects of APS collagen sponge on enhancing angiogenesis and collagen synthesis so as to provide evidence for the future tissue engineering application as a kind of angiogenic scaffold.Methods APS collagen sponges were prepared by covalent binding with collagen polypeptides by using of crosslinking agents at the ratio of 1 ∶ 1(W/W).Twenty 10-week-old Sprague Dawley rats(10 males and 10 females,and weighing 200-250 g) were selected.Longitudinal incision was made at both sides of the back to form subcutaneous pockets.APS collagen sponges of 5 mm × 5 mm × 5 mm at size were implanted into the left pockets as the experimental group,collagen sponges without APS of the same size into the right pockets as the control group.The general conditions were observed after operation.At 3,7,14,and 21 days,5 rats were sacrificed and the samples were harvested to count the number of microvessels,to measure the contents of the hydroxyproline(Hyp),and to detect the mRNA expressions of angiopoetin 1(Ang1),matrix metalloproteinases 9(MMP-9),and tissue inhibitors of metalloproteinases 1(TIMP-1).Results All rats were alive during experiment period.The number of microvessels increased gradually,and reached the peak at 14 days in 2 groups;the expermental group was significantly higher than the control group(P 0.05).The contents of Hyp increased gradually in 2 groups,and the experimental group was significantly higher than the control group(P 0.05).The mRNA expressions of Ang1 and MMP-9 in the experimental group were significantly higher than those in the control group at 3,7,and 14 days(P 0.05);the mRNA expression of TIMP-1 in the experimental group was significantly lower than that in the control group at 3 days and was significantly higher at 14 and 21 days(P 0.05).Conclusion The APS collagen sponges can improve angiogenesis and collagen synthesis in wound healing by regulating the expressions of Ang
